An Experimental Study On Blastocyst Biopsy For Preimplantion Genetic Diagnosis And The Mouse Blastocyst Vitrification

Objective (1) Try to estimate the value when use the kidney-strengthing herbs— Yangjing Zhongyu decoction(YJZYD) to reinforce the superovulation on mouse and improve the embryonic development potentiality. (2) Discuss the feasibility by catch trophectoderm cells through blastcoelecentesis biopsy; Observe the blastocysts growth after the centesis and attempted use those trophoblastic cells to complete the karyotype analysis and whole genome amplification,stand groundwork for preimplantion genetic diagnosis . (3) Observe the vitrificate effect after balstocysts punctured ,try to see if use freezed plats and add serum substitute supplement into cryoprotective agent can improve it.In short,by this animal experimental study,we want to find out a new effe ctive and convenient way for preimplantion genetic diagnosis and improve some technique on blastocyst vitrification.Methods 200 healthy female Kunbai-mice and 40 male mice were used for the experiment.Intragastral administrated YJZYD to femal mice,observe the change of the estrous cycles and record the number of the oocytes after superovulation ,take those in vitro fertilization and record growth of those embryos. The trophoblastic cells were pumped by the micromanipulation apparatus from blastocoel and used for the karyotype analysis and primerextension preamplification-nested PCR..Then transferred to the uteri of pseudopregnant surrogate mother mice and wait for the babies. In other hand, add the SSS into cryoprotectant and use the freezed plats to cryopreservate some blastocysts.Result 1 .After taken YJZYT for two weeks,the estrous cycles in mice were shorten and inerratic,the average time was 4.84 days, the average number of oocyte-obtained...
