objective: To study if the vector expressing HPV-16 E6 siRNA can inhibit expression of HPV-16 E6 gene in CaSki cell line of cervix cancer, and to learn the effect of inhibiting E6 gene expression in CaSki cell line.Methods: To construct vectors expressing small inference RNA of human papillomavirus 16 E6 gene, and transfect these vectors into human cervic cancer CaSki cell line, detect expression of E6 mRNA by RT-PCR and expression of P53 protein by western blot. Analysis cell cycle of CaSki cell line by flow cytometry and observe the apoptosis of the cells.Results: After transfected the vectors expression HPV-16 E6 siRNA into CaSki cell line, the expression of HPV-16 E6 mRNA was down-regulated and the expression of P53 up-regulated in CaSki cell line, and the cell cycle arrest at G0-G1 stage. The proliferation of CaSki cell was inhibited and the cell was induced apoptosis.Conclusion: RNAi can silence the expression of HPV-16 E6 gene that integrate into CaSki cell genome, which up-regulate expression of tumor suppression protein P53 and result in CaSki cell growth retardation and induce apoptosis of CaSki cell.
