Effect Of Rosiglitazone On Diethystilbestrol (DES)-induced Rat Prolactinoma

Objectives:To develop a prolactinoma model induced by Diethystilbestrol(DES) in femal Wistar rats and study the effects of rosiglitazone on rat prolactinoma.Methods:Firstly, twenty femal Wistar rats were divided into 2 groups at random. Each rat in A0 group(DES group):12,was subscutaneously implanted with DES encupsulated in silastic tubing. The rats in B0 group(control group):8, were implanted with a blank silastic tubing. After 8 weeks, all the animals were executed to investigate the weight of pituitary gland, serum prolactin(PRL) level, pituitary light microscope, PRL immunoreactive cell countings and CyclinD1 expression in prolactinoma. Secondly, forty female Wistar rats were subscutaneously implanted with DES encapsulated in silastic tubing. After 8 weeks, rats were diveded into 5 groups at random. A, control group; B, bromocriptine group; C, high dose rosiglitazone group; D, middle dose rosiglitazone group; E, low dose rosiglitazone group . After 4 weeks of treatment all the animals were executed to investigate the weight of pituitary gland, serum PRL level, pituitary light microscope, PRL immunoreactive cell countings and CyclinD1 expression in prolactinoma.Results:Subscutaneous DES implantation is capable of forming prolactinoma in female Wistar rats. After 8 weeks, the serum PRL levels, weight of pituitary gland, PRL immunoreactive cell countings, were significantly lower than those in A,D;E or A0 group after treatment with bromocriptine in B group and rosiglistazone in C group(P<0.05). All the parameters mentioned above were no difference between B and C group(P>0.05). The expression of CyclinD1 in C group were significantly lower than that in A,B or A0 group(P<0.05).Conclusion:Rosiglistazone ( 20mg.kg^-1.d^-1 ) has effect against DES-induced rat prolactinoma.
