Backgrounds:Multiple myeloma is a clone disease with a feature of low proliferation and low apoptosis.It is a worrying disease for low remission rate and high relapse rate.It can not be cured with conventional therapies.The patients lived a poor quality and transient life.The conventional therapies can not prolong the period of the disease free survival (DFS) and overall survival (OS) presently .The minimal residual disease(MRD) are always the major causes for MM resistant to drugs and relapse. Therefore, new therapeutic techniques are needed for combating this disease. Now people desire to eradictate MRD by immunotherapy after chemotherapy. Immunotherapy for malignant tumors has been developed rapidly since 1980'.Recent studies have focused on the adoptive immunotherapy for malignancies mediated by cytotoxic lymphocyte (CTLs).MM is suitable for immunotherapy because of the expressing of tumor specific antigen and tumor associated antigen. Granulocyte- macrophage colony stimulating factor(GM-CSF) as an importat cytokine for antigen presenting cell(APC),has the capacity to keep maturation of DCs ,up-regulate their antigen captureing and enhance the surface expression of costimulatory molecules.So we plan to induce MM U266 specific CTLs mediated by DCs in vitro, which pulsed with U266 cell line antigens and transfected with recombinant adenovirus vector containing human GM-CSF, and assess the lethal effect of cytotoxic lymphocyte against U266 cells.This study will set up a base for the gene therapy and immunotherapy of MM.Objective: The goal of the study was to assess the lethal effect of cytotoxic lymphocyte against U266 cells induced by DCs pulsed with MM U266 lysate and transfected with GM-CSF recombinant adenovirus.Methods:The PBMCs were isolated by Ficoll-paque and monocytes were received by...
