Objective:To establish the rat model of experimental autoimmune encephalomyelitis (EAE), andcalculate the CD4~+ T lymphocytes in peripheral blood and central nervoussystem(CNS) of EAE rats, and observe the effect of triptolide on CD4~+T lymphocytesin peripheral blood and central nervous system, aim to provide experimental evidencefor clinical application.Methods:The EAE model was induced in Wistar rats by injecting four foot pads withguinea-pig spinal cord homogenate in complete Freund's adjuvant (GPSCH-CFA),and coinjecting one foot pad with bordetella pertussis vaccine(BPV). Rats wererandomly divided into EAE group and control group, and each group was thenrandomly divided into medication administration group and normal saline controlgroup. Rats were weighed, and clinical scores were evaluated every day; Thehistomorphological changes were investigated by HE staining; The amounts of CD4~+T lymphocytes in peripheral blood were calculated by flow cytometry; The CD4~+Tlymphocytes within the central nervous system were investigated byimmunohistochemistry.Results:1 .The EAE model had been successfully established, the clinical symptoms of ratsoccurred at about 11 days post-immunization, and reached the peak at 17 dayspost-immunization, the body weight decreased obviously, and the infiltratedinflammatory cells within the axion were considerable.2.
