Objective:1 To establish the rat model of experimental autoimmune encephalomyelitis (EAE)2 To observe the therapeutic effect of triptolide (Tri) on EAE3 To observe if triptolide can inhibit the infiltration of inflammatory cell in the central nervous system (CNS) of EAE rat model, and decrease the inflammatory reaction4 To observe the effect of triptolide on matrix metalloproteinase-9 (MMP-9) expression in the CNS of EAE rats5 to seek a new consider to therapia EAE and multiple sclerosis (MS) which has extensively potentialMethods:Wistar rats were randomly divided into EAE group and complete Freund's adjuvant (CFA) group.The EAE group was induced by injecting four foot pads with guinea-pig spinal cord homogenate in complete Freund's adjuvant(GPSCH-CFA) 0.5 milliliter, and coinjecting one foot pad with bordetella pertussis vaccine(BPV) 0.1 milliliter every rat. The CFA group was injected with CFA and NS. Each group was then randomly divided into medication administration group for triptolide and control group (isovolumic normal saline). Rats were weighed, and clinical scores were evaluated every day; The histomorphological changes were investigated by HE staining; observe the MMP-9 expression in the central nervous system of EAE rats by immunohistochemistry.Results:1.The EAE model had been successfully established, the attack rate was 91.6%. the clinical symptoms of rats occurred generally at about 11 days post-immunization, and reached the peak at 16 days post-immunization.But none of the CFA groop was attacked. the body weight decrease and the attack rate of EAE/Tri subgroop was less than EAE/NS subgroop,. The max clinical scores was obviously lower also. They were all statistical significance (P<0.05) .2.In HE staining the amount of inflammatory cell infiltrated the brain and spinal cord of the EAE/Tri subgroop.It was lower in EAE/NS subgroop.3. The MMP-9 expression of the EAE/Tri subgroop in the central nervous system was lower then EAE/NS subgroop.Conclusion:1. The EAE model had been successfully established by injected with GPSCH- CFA and coinjected with bordetella pertussis vaccines (BPV).2. There were considerable inflammatory cell infiltrated in the CNS of EAE rats.3.Triptolide could reduced the attack rate of EAE obviously.4. Triptolide may reduce the MMP-9 expression in the central nervous system of EAE rats and suppress the attack of EAE, which suggest that triptolide has promising perspective in MS therapy in mankind.
