Objective: To observe on acute toxic effects in mice after different doses of the toluene intraperioneal injection just as follows: â‘ the general and the toxic phenomenon of nervous system and other organs in mice ;â‘¡the pathologic changes of the different organs, such as: liver/ heart/ kidney/lung ;â‘¢the changes of the neurotransmitters and the enzyme owing to the acute toxicity using immunohistochemistry methods;â‘£and explored the acute toxic mechanisms. Methods: To make the acute toxic mice models of intraperioneal cavity injection , the toluene groups could be classified into five teams:8000mg/kg ,4000mg/kg ,2000mg/kg ,1000mg/kg ,500mg/kg. The control group was normal sodium: the number of each team was 15, of which the male and female numbers were equal. After using toluene, the acute toxic performances and record and number of death were observed, whose hepatic tissues were used to analyze the liver ratio of wet weight so that the hepatic injuries could be described quantitatively. The programmatic controlling autonomic activities case could be used to detect and record the activities in 3min. The influences of toluene acute toxicity on mice autonomic activities could also be known. All these operations were carried out in 4d,6d,8d,12d,14d respectively. Four groups: 2000mg/kg, 1000mg/kg, 500mg/kg and normal sodium could be selected . The brain ,liver, heart, kidney, lung, tissues in ld,7d,14d were used to make paraffin imbedding and section using the tissues originated from the brain, liver, heart, kidney and lung of mice as well as the same tissues from the death. These processes were just as followsâ‘ To observe the historical and pharmacological changes after normal HE. â‘¡To observe the changes of cerebral cortex and Nissl bodies in the neurons of HIP after brain tissue Nissl. â‘¢ To use immunohistochemistry ABC method to explore the changes of the ChAT, GABA, Glu positive cells in the mice cerebral cortex.
