Objective:To observe the acute toxic effects on heart and liver of mice.Methods: To make the acute toxic mice moels of intraperioneal cavity injection, the mice were randomly divided into four groups, each group of 48 mice, and they were equally divided between male and female. The control groupws normal sodium(800mg/kg) , the toluene groups could be classified into three teams:low-dose group (400mg/kg), medium-dose group(800mg/kg), high-dose group(1200mg/kg).①To observe its behavior after using toluene;②The mice were sacrificed when the time is 3h,6h,12h,24h,2d,3d,4d,5d,whose hepatic and heart tissues were used to analyze the liver and heart ratio of wet weight so that their injuries could be desribed quantitatively;③In the above-mentioned time point, use blood samples to detection the creatine kinaseisoenzyme (CK-MB), creatine kinase (CK), aspartate transferase (AST), lactate dehydrogenase (LDH) activity;④In the above time spot, take the partial liver organization to make the tissue homogenate, determination the acyivity of superoxide dismutase(SOD) and malondialdehyde(MDA);⑤Each group takes six mice to examination electrocardiogram when time is 30minutes;⑥The liver and heart tissues used to make paraffin imbedding,in order to observe the historical and pharmacological changes after normal HE.Results:①The team of using toluene all showed acute toxic performances such as inhibitory state after short excited,dull reaction et al, the mice body weight drops near to 1g in 24 hours;②Acute toluene exposure have impact on heart coefficient. The control group had significant difference (P <0.05) with 24h,2d,4d in low-dose grope, 24h,2din middle-dose grope, 24h,2d,3d,4d,5d in high-dose grope,while the coefficient of the liver was less affected, 6h in low dose group, 3h in medium dose group compared with the control group have significant difference (P <0.05);③The activity of CK-MB ,CK, AST and LDH showed a certain correlation with toluene dose,that is the same time the higher dose causes the enzyme activity markup to be more obvious; while at the same dose group,the activity of myocardial enzyme were dropping with timeisdecreasing;④Acute toluene exposure caused SOD and MDA has a certain degree of change. Much time spots in each group had significant difference (P <0.05) with the control group;⑤Each group exposure in toluene which J-point apparent displacement(P﹤0.05);⑥HE:Heart muscle cellsswelling and cardiac muscle fibers,partially dissolved;.Liver mesenchymal dropsy,and its rope disorder,with increasing doses of toluene,we can see its nuclear fragmentation and dissolution.Conclusion:①The toxic effects of toluene on the mice were found after intraperioneal injection, the heart and liver of the mice are all experiencing varying degrees of injury;②After acute toluene exposure the mice showed independent activity after a short excited state,and the weight assumes the declining trend.
