Object: The development of a kind of hydrogel dressing with a simple technological process,premium properties and good effectiveness to protect wound from infection is necessary for the promotion of wound healing. In this study,two kinds of hydrogels'properties and their rules of in-vitro drug delivery were observed. And the results of this study can provide a experimental basis for the use of hydrogels in treating wound infection. Meanwhile,an animal model of radiation burn was established successfully,which is not only helpful to make certain of the secretion regulation for endogenous VEGF,but also lays the foundation for further research on exogenous VEGF's effect on the therapy for radiation burn.Methods: Effect of the irradiation and the physical management on potency of the gentamicin was evaluated by diffusion method. The hydrogel films containing drug or not were prepared by radiation crosslinking technique or physical crosslinking technique and their hydrating and evaporation properties,swelling properties,mechanical strength and the rules of the durg release in vitro were studied. And the content of VEGF in rats, serum was detected by ELISA in the study,too.Results: Potency of the gentamicin irradiated to 37.5 kGy and treated by the physical management did not change. The radiation crosslinking hydrogel showed favourable water-absorbing property. D hydrogel displayed eligible degree of swelling and satisfactory mechanical properties. The drug delivery of the two hydrogels both conformed to Higuchi kinetic equation, and mechanisms of the drug release were matrix diffusion. The level of VEGF protein in rats'serum was not detected.Conclusion: The results showed the two kinds of hydrogels had satisfactory properties and could control the drug release,which may value to give evidence for wound infection healing. The establishment of animal model of radiation burn provides a experimental basis for exogenous VEGF's effect on the therapy for radiation burn.
