Study On Preparation And Analysis Of Ginsenoside-Rg3,-Rh2

Ginseng ,the root of of Panax ginseng C.A.Mey of the Araliaxeae family. It is one of the most widely used Chinese traditional medicines .The ginsenosides are the main active components of ginseng. Recently studies on antitumor action of several rare ginsenosides, especially ginsenoside-Rg3 and -Rh2 ,were attracting .The preparation and analysis of ginsenoside -Rg3 . -Rh2 were studied in this paper.Some traditional preparation methods of ginsenoside -Rg3 . -Rh2 have disadvantages such as long times ,microwaves can speed up chemical reaction, preparation of ginsenoside-Rg3 and -Rh2 from the ginsenosides of protopanaxdiol group by hydrolysis with microwaves method was carried out. The optimal conditions for ginsenoside -Rg3 was 0.5mol/L of HC1;110℃; 15 minute of reaction time. The optimal conditions for ginsenoside -Rh2 was lmol/L of HC1;120℃; 20 minutes of reaction time.Simultaneously a new analysis method-HPLC determination of ginsenoside-Rg3 and -Rh2 was been established in this paper .The optimal HPLC conditions was as follow : column was Agilent ZOBAX Eclipse XDB C18;The wavelength for detection was 203 nm. mobile phase was aacetonitrile:water=1:1(v:v),The flow rate was 1ml/min.Under these conditions,a baseline separation of ginsenoside -Rg3 -Rh2 was obtained and the linear rang of ginsenoside -Rg3(S) is 0.083ug/ul to 0.664ug/ul,the related coefficient is 0.9996,the average recovery is 98.9%. the linear rang of ginsenoside -Rg3(R) is 0.189ug/ul to 3.024ug/ul,the related coefficient is 0.9996,the average recovery is 97.7%. the linear rang of ginsenoside -Rh₂(S) is 0.289ug/ul to 2.312ug/ul,the related coefficient is 0.9995,the average recovery is 98.5%. the linear rang of ginsenoside -Rh₂(R) is 0.130ug/ul to 1.044ug/ul,the related coefficient is 0.9998,the average recovery is 98.4%. The results showed that this method was brief and reliable.
