| Objective:To examine the expression of hemeoxygenase–1(HO–1) and hemeoxygenase–2(HO–2) in placenta tissue and the level of its byproduct carbon monoxide(CO) in peripheral blood from patients with hypertensive disorder complicating pregnancy and women with normotentive late pregnancy , so as to explore the role of HO–CO system in the pathogenesis of hypertensive disorder complicating pregnancy.Methods: We examined the expression and localization of HO-1 and HO-2 in placenta tissue of hypertensive disorder complicating pregnancy (study group,n=30)and normotentive late pregnancy (control group,n=26) using in situ hybridization and immunohistochemistry. Furthermore ,medical–image measure system was used for quantitative analysis. In addition , the level of CO–hemoglobin(COHb) in peripheral blood was studied.RESULTS:1. HO–1 and HO–2 were expressed in placenta tissue of hypertensive disorder complicating pregnancy and normotentive late pregnancy group . HO–1 is expressed prominently in placental trophoblast cell, HO–2 prominently in trophoblast and villus vascular endothelium cell.â‘ In situ hybridization method demonstrated that the average optical density of HO–1mRNA in cell of trophoblast and HO–2 mRNA in cell of trophoblast and villus vascular endothelium were(0.260±0.032),( 0.258±0.025)and(0.252±0.031) in hypertensive disorder complicating pregnancy group,which had no significant difference compared with those in control group(0.260±0.026),(0.265±0.030)and(0.269±0.037) (P>0.05).â‘¡Immunohistochemical method demonstrated that the average optical density of HO–1 protein in cell of trophoblast(0.243±0.034) were significantly reduced in placenta from hypertensive disorder complicating pregnancy, compared with normotentive late pregnancy group(0.266±0.033)ï¹P<0.05﹞. The average optical density of HO–2 protein in cell of trophoblast and villus vascular endothelium(0.240±0.035)and(0.242±0.034) were significantly lower than those in control group (0.261±0.040)and(0.267±0.036)ï¹P<0.05﹞.
