Chronic fatigue syndrome (CFS) is a kind of syndrome that profoundly manifested in long term of extreme exhaustion, which occurs mostly among women at the age of 20 to 50.It belongs to the category of sub-health, which concerns mostly about the human health in the 21st century. It belongs to deficiency and exhaustion, insomnia, palpitation, lily disease in traditional Chinese medicine (TCM). As the pathogenic factor and etiopathogenesis of the disease is unclear up till now, there are not effective measures for prevention and treatment. Research of modern medicine on CFS mainly manifests in its etiopathogenesis and the method of model preparation .It is commonly accepted that CFS is related with long-term spiritual stimulation ,excessive exhaustion of brain and physical force ,and virus infection, accompanying with malfunction of nerve-endocrine-immune system .There are two methods in model preparation: one is "chronic bondage", another is the complex of chronic bondage and swimming in cold water, accompanying with electrical stimulation while eating . According to the above two methods and learn from other's experience, this experiment adopted the method of chronic bondage and swimming in the cold water alternately in model preparation, which proved to be reasonable and convenient.Acupuncture and moxibustion is a treatment that regulates the function of the neural and endocrine system of the body and reinforces the immune function and the capability of preventing diseases. It is prior in wholly regulation, and has the advantage of easy manipulation and convenience, no side effect, being economical, effective and safe. As one method of acupuncture and moxibustion, acupoint cat gut imbedding has the characteristics being convenient, time-saving, soft and durable, so it has amplitude foreground in preventing and treating diseases. So that, on the basis of literature research, this thesis demonstrated the theory foundation of acupuncture and moxibustion treating CFS. More over, designed and carried out the research of cat gut imbedding regulating the neural and endocrine function of chronic fatigue rats. On observing the change of weigh and behavior and the index of monoamine and glucocorticoid both in centrum and peripheral, we try to find out the theory of cat gut imbedding regulating the endocrine function of chronic fatigue rats.1. MethodRandomly divided the animals into model group, cat gut embedding group, ginsenoside group and normal control group. We got the rat model of chronic fatigue by the method of chronic bondage and swimming in cold water alternately .The cat gut embedding group used the acupoints of Housanli, Baihui, Shenshu according to Experimental Acupuncture Science, alternately on both sides. Ginsenoside group was stomach fulfilled by ginsenoside. The normal control group was not treated. Four weeks later, measured the behavior of rats, recorded the weigh of the rats, then had their head cutted and blood sampled. Recorded the weigh of the hypothalamus and pituitary, then measured NE, DA, 5-HT, ACTH and CORT.2. Results(1) Weigh: As time goes, the model group ate less and drank less. Their hair lost luster and they lost weigh obviously. There is remarkable difference compared with the normal control group. (PO.01);while the cat gut embedding group gained weigh more than the model group, and there are statistics difference (P< 0.05).(2) Change of behavior: the measure of open-field activity showed that, the model group had distinctive decrease in horizontal and vertical motion when compared with the normal control group (P<0.01). The experiment of tail-hanging showed that, the still period after tail-hanging of the model group was obviously longer than the other groups;The still period of the cat gut embedding group was decreased, and the difference was distinctive compared with the model group.(3) ACTH and CORT in plasma: the model group was obviously higher than the normal control group and ginsenoside group ( P< 0.01).The cat gut embedding group had statistics difference compared with the normal control group and model group ( P <0.05 and PO.01 respectively).(4) Mono-amine neural transmitter in the hypothalamus and pituitary: NE: the model group ascended remarkably and had distinctive difference with the normal control group (P< 0.01). The difference is statistic when compared with the cat gut embedding group and the ginsenoside group. 5-TH: The model group descended remarkably. The difference is distinctively when compared with the normal control group (P < 0.01) and statistic with the cat gut embedding and ginsenoside group (P <0.01). DA: the model group, cat gut embedding group, and ginsenoside group had slight raise as compared with the normal control group, the raise of the model group was higher but no statistic difference (P> 0.05).(5) Mono-amine neural transmitter in plasma : NE : the model group descended obviously , the difference was distinctive with the normal control group ( P< 0.01 ) and was statistic with the cat gut embedding and ginsenoside group ( P< 0.05 ) . 5-HT: the model group rose obviously, the difference was distinctive with the normalcontrol group (P< 0.01) and was statistic with the cat gut embedding and ginsenoside (P< 0.05).DA: the model group, cat gut embedding and ginsenoside group descended slightly, but no statistics difference with the normal control group (P< 0.05). 3. Discussions(1) Cat gut embedding increased the weigh of model rats, elevated their emotional responses and seeking activities, which proved that cat gut embedding, improved the physical force of chronic fatigue rats and excitability of their neural system, increased their spontaneous activity.(2) Cat gut embedding decreased the content of glucocorticoid in plasma of model rats, suggesting its function in resisting the capability of responding to stimuli and fatigue may be related with its inhibition to the cortex of adrenal gland.(3) Cat gut embedding regulated the disorders of mono-amine neural substance in the hypothalamus, pituitary and peripheral of model rats, which indicated that regulating the excitability of hypothalamus- pituitary -adrenal axes (HPA) and regulating the disorders of mono-amine neural substance may be the effective mechanism of cat gut embedding.
