Studies show that Rhodiola has a good effect on fatigue, but active ingredients with anti-fatigue effects and the mechanism are not yet clear. Brain tissue of exercise-induced fatighue rats were studied inorder to study the effcts of Rhodiola glycosides on the monoamine neurotransmitter of central nervous system in exercise-induced fatigue rats. And at the same time, based on this, the mechanism of anti-fatigue effects was invited so as to provide a scientific basis for the study of the anti-fatigue mechanism of Rhodiola glycosides and the development of new anti-fatigue drugs.Centrol fatigue model was invited for the first based on the 10d swimming with increasing force and the result showed that after 10 d of incremental swimming training, rats showed alopecia, tired, decreased appetite, decreased exercise capacity, sluggish response to external stimuli, and such characteristics of depression, and exploratory behavior decreased, body weight growth.Effects of anti-fatigue was studied by using of the swimming test and the result showed that, compared with the negatve control group (swimming time 30.5min), each dose of Rhodiola glycosides could prolong the swimmig time signnificantly. And it indicated that Rhodiola glycosides could enhance the endurance of exercise-induced fatigue rats.5-HT,5-HT, DA and NE levels in the exercise-induced fatigue rats were detectived by using of high performance liquid electrochemical method.Reults showed that,5-HT could be better separated, the retention time was10.3min.There was a od linear relationship between concentration and peak area. The regression equation and correlation coefficients were:y= 5179.1x-4124.3, R2= 0.9964.5-HIAA, DA and NE could also be well seperated.Their concentations were all in 1-10μg/g and the concentration and peak area got a good linear relationship. For 5-HIAA,the retetion time was 22.94 and the regression equation and correlation coefficient were:y= 14075x-9472.3, R2= 0.9954. For DA,the retetion time was 5.448 and the regression equation and correlation coefficient were:y=4086.9x-4398.7, R2=0.9957. For NE,the retetion time was 3.729 and the regression equation and correlation coefficient were:y=5179.1x-4124.3, R2=0.9964.The results showed that, compared with the negative control group,5-HT in each treatment group were significantly lower (P<0.05), and with the increased dose of Rhodiola glycosides the content decreased significantly. With increase of the recovery time,5-HT levels in the treatment group decreased in 6-24h, while the negative control group showed an increasing trend.5-HIAA in each treatment group were significantly lower than the negative control group, and the difference was statistically significant (P<0.05). And with the increase doses of Rhodiola glycosides the content decreased significantly. With increase of the recovery time,5-HIAA levels in the treatment group decreased in 6-24h, while the negative control group showed an increasing trend. The DA treatment group was significantly higher than the negative control group, and the difference was statistically significant (P<0.05). With the increase of recovery time, DA levels in the treatment group increased at 6h-24h recovery period, while the negative control group showed an decreasing trend. NE content in the treated group was significantly higher than the negative control group, and the difference was statistically significant (P<0.05).Wiith the increase doses of Rhodiola glycosides,NE content increased significantly. And with the increase of the recovery time, NE levels in the treated groups increased at 6-24h, while the negative control group showed a trend of decreased first and then increased. In conclusion, Rhodiola glycosides could prolong the swimming time, and it had a significant anti-fatigue effect. Rhodiola glycosides could significantly reduce the inhibitory neurotransmitter, delay the effects of the inhibitory neurotransmitter, while improve the content of the excitatory neurotransmitter, and enhance athletic ability, and thus play the role of anti-fatigue.
