1-deoxynojirimycin (DNJ) and its ramifications are effectiveα- glucosidase inhibitors, they are used in curing diabetes, fatness and virus infections. Today DNJ has been found in many kinds of plants and microbes especially in mulberry leaves. The silkworm feeds on mulberry leaves during all its life, it can accumulate DNJ, the content of DNJ in its body is much higher than in mulberry leaves. So DNJ is an important physiological active material in mulberry leaves and silkworm body, especially when related in its reducing blood glucose. However, restricted by the detecting methods, at present, there is no systematic research about the change and accumulation law in mulberry leaves and silkworm body. According this, this article develops a new method in detecting DNJ, which is RP-HPLC-UV. Through this new detecting methods, we have investigated the change law of DNJ both in mulberry leaves and in silkworm body, and discussed its effect in reducing blood glucose, so as to find out the high DNJ content silkworm and mulberry resources and some other biology resources, and establish a quality guideline on the reducing effect blood glucose of silkworm powder and mulberry leaves. the main research findings are as follows.1 DNJ was extracted from silkworm and mulberry leaves powder by using 0.05mol/L HCl. after blending DNJ stand solution or silkworm powder or mulberry leaves powder extraction and borate potassium solution (pH= 8.5), FMOC-Cl was added to derive DNJ. it can produce the DNJ-FMOC which has the ability to absorb ultraviolet radiation. Through the findings we find that the separation chromatogram of the production is very satisfactory. The peak area of DNJ-FMOC is respondent to the consistency of DNJ. the average of recovery rate is 100.7%.the precision test, stability test, recurring test and the recycling test all shows that this method is credible and stable, and it can be used as an effective method in detecting the DNJ content in mulberry leaves, silkworm and other medicines.2 Using RP-HPLC-UV method to detect the DNJ content in different silkworm powders, and find that the DNJ content is influenced by silkworm...
