[OBJECTIVE] To study the effects of serums from SD rats fed with Yunnan-Baiyao on the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) from SD rats and the mechanism of yunnan-baiyao in promoting the healing of bone fracture.[METHODS] BMSCs were identified by their morphology analysis and their capacities in the osteogenic, chondrogenic and adipogenic differentiation. The serums from SD rats fed with Yunnan-Baiyao were abtained by intragastric administration of Yunnan-Baiyao . The proliferation of BMSCs were evaluated via MTT assay.The osteogenic differentiation markers were determined by para position Nitrobenzene phosphoric acid (pNPP) and Alizarin Red S staining.The expressions of CBFA1 and VEGF mRNA in BMSCs were detected by RT-PCR.[RESULTS]Following treatment with the medium containing 5%-10% serums of rats fed with yunnan-baiyao from dl to d7,no significant effect in the proliferation of P2 BMSCs was observed(.P > 0.05).After 7 days treatment with 10% serums from rats fed with Yunnan-Baiyao,the ALP activeties of P2 BMSCs were increased obviously and were higher than that in the control groups(P <0.01); the mRNA expressions of CBFA1 were 1.26 fold higher than that in the control groups(P<0.01),After 14 days treatment, the ALP activeties in the Yunnan-Baiyao groups were 1.45 fold higher and the mRNA expressions of VEGF were 1.38 fold higher than that in the control groups (P <0.01).After 21 days treatment ,the quantities of calcium nodule in BMSCs were 4.41 fold higher than that in the control groups(P <0.01). [CONCLUSION] The serums from SD rats fed with Yunnan-Baiyao had no obviously effect on the proliferation of P2 BMSCs,but they could enhance differentiate into osteoblasts.They could increase the expressing of CBFA1 and VEGF mRNA. Therefore,one of the possible mechanism of Yunnan-Baiyao in promoting bone healing was to enhance the osteogenic differentiation of BMSCs and induce the angiogenesis in the tissues around bone fracture.
