[Abstract] Objective 1. To study whether or not the Chinese patients with gastric cancer and patients with duodenal ulcer are associated with TNF-βgene polymorphisms. 2. To study whether or not the Chinese patients with gastric cancer and patients with duodenal ulcer are associated with IL-4 gene polymorphisms. 3. To analyze the H. pylori serologic marker among gastric cancer patients, duodenal ulcer patients and control subjects. 4. To study the co-effects of TNF-β,IL-4 gene polymorphisms and H. pylori infection in the development of gastric cancer and duodenal ulcer.Methods Subjects were divided into three groups. 151 gastric cancer patients,109 duodenal ulcer patients and 107 control subjects were enrolled. Patients with gastric cancer were recruited by pathological diagnosis. Patients with duodenal ulcer were excluded NSAIDs administration. Patients with control subjects were excluded peptic ulcer history. TNF-βpolymorphism was analyzed by PCR-RFLP. IL-4 gene polymorphism was analyzed by PCR-SSP. Serum H. pylori IgG was tested by ELISA. The results were analyzed by SPSS13.0.Results 1. The gene frequency of allele A of TNF-β+252 in gastric cancer patients (50.0%), especially in noncardia gastric cancer patients(51.5%), was significantly higher than that in control subjects(39.7%)(P<0.05). The OR estimates for TNF-β+252*A were 1.519(95%CI 1.054~2.187)for gastric cancer, and 1.612(95%CI 1.087~2.392)for noncardia gastric cancer. The TNF-β+252A/A homozygote was significantly predominant in patients with noncardia gastric cancer (OR=2.304,95%CI 1.163~4.565), whereas there was not significantly correlated with cardia cancer. There was also no significant difference in the gene frequency of allele A between duodenal ulcer and control subjects(P>0.05). 2.
