The Association Of The TNF, Interleukin-10 Gene Polymorphisms With Gastric Cancer

PartⅠ:The relationship among TNF gene polymorphisms, Helicobacter pylori infection and gastric cancerObjective To study the distribution of TNF gene polymorphisms in patients with gastric cancer in Hubei province with Han nationality and their association with Helicobacter pylori (Hp) infection.Methods 605 patients with gastroduodenal diseases (196 chronic gastritis,189 gastroduodenal ulcer and 220 gastric cancer) and 624 healthy controls were genotyped by the PCR-RFLP method for TNF-α308, lymphotoxin-α(LT-α) Nco I and AspH I gene polymorphisms; TNFd microsatellite alleles were amplified by allele-specific PCR. The amplified products were electrophoresed with 12% polyacrylamide gel, followed by silver staining. At the same time, the PCR products were cloned and sequenced. Hp infection status was determined by ELISA.Results①Frequency of LT-αNco I AG genotype in patients with gastric cancer with Hp infection was significantly higher than that in Hp positive healthy controls (P< 0.05). There were no other associations between TNF-α308, LT-αNco I and AspH I gene polymorphisms and Hp infection in gastroduodenal diseases.②Eight alleles at the TNFd locus were detected. Statistical analysis showed that no difference was observed between patients and healthy controls in the distribution of alleles at the TNFd loci (P>0.05). The frequence of d2/d6 genotype was significantly higher in patients with gastric cancer than in healthy individuals (P<0.05); The frequence of d3/d4 genotype was significantly lower in patients with gastric cancer than in healthy individuals (P< 0.05). d2/d6, d3/d4 genotype wasn’t related to age or gender or grade of differentiation or clinicopathologic stage in gastric cancer.Conclusions①LT-αNco I AG heterozygous genotype could be associated with Hp infection in patients with gastric cancer in Hubei province with Han nationality.②d2/d6 and d3/d4 genotypes are associated with the susceptibility to gastric cancer.PartⅡThe relationship among IL-10 gene polymorphisms, Helicobacter pylori infection and gastric cancerObjective To study the distribution of IL-10 gene polymorphisms in patients with gastric cancer in Hubei province with Han nationality and their association with Helicobacter Pylori (Hp) infection.Methods 605 patients with gastroduodenal diseases (196 chronic gastritis,189 gastroduodenal ulcer and 220 gastric cancer) and 624 healthy controls were genotyped by the PCR-RFLP method for IL-10 -1082,-819,-592 gene polymorphisms. The genotype frequencies of IL-10-819 were same as IL-10-592.③The frequency of IL-10 -1082 AG+GG genotype with Hp positive of gastric cancer patients was significantly higher than that of control groups (P< 0.05).Conclusions①Allele AG+GG of IL-10-1082 was associated with gastric cancer in Hubei province.②There was associations between IL-10- 1082 AG+GG and Hp infection of gastric cancer.
