Purpose: To study the growth inhibitory action and inducing apoptosis effect of Nacoscale realgar, Qing dai and Compound huang dai on Acute promyelocytic leukemia (APL) cell strain NB4 cell, and to provide the evidence about the mechanism of Compound huang dai treating APL and its main effective ingredients.Methods: This experiment studied Compound huang dai by resolution. APL cell strain NB4 cell was used as vitro model. There were four groups including Nacoscale realgar, Qing dai, Compound huang dai and blank in this experiment. This experiment included two parts:(1) Methylthiazoletrazolium(MTT)assay detected the growth inhibitory degrees afterdrugs treated NB4 cell:At first, the final concentration of Nacoscale realgar ranged from 0.1875 mg·L-1 to 12 mg·L-1, the final concentration of Qing dai did from 0.235 mg·L-1 to 15.04 mg·L-1 and the final concentration of Compound huang dai did from 0.656 mg·L-1 to 42 mg·L-1. These drugs treated NB4 cell respectively, after 24 hours MTT assay was used to detect the growth inhibitory degrees. Then, the correlation of logarithm of drug concentration to the inhibitory rates was analyzed with linear regression and linear equation was obtained, from which the half inhibitory concentration (IC50) came out. Finally compare the growth inhibitory rates of three experimental groups.Then, 1.5 mg·L-1 Nacoscale realgar, 1.88 mg·L-1 Qing dai and 5.25 mg·L-1 Compound huang dai treated NB4 cell respectively and after 6 hours, 12 hours, 24 hours, 48 hours and 72 hours MTT assay was used to detect the growth inhibitory degrees.
