Objective: To investigate and testify the protective effect of puerarin and its mechanism on transient focal cerebral ischemia.Methods: Eighty SD rats were randomly divided into 5 groups: A group (sham operation group), B group (ischemia control group), C group(lower dose (75mg/kg) puerarin group), D group (higher dose (150mg/kg) puerarin group) and E group (troxerutin group) with 16 rats in each group.①Build model: Focal cerebral ischemia models in middle cerebral artery were made with threaded method.The blood stream in middle cerebral artery was blocked for 2 hours and then reperfusion for 24 hours.The rats in the sham operation group didn't embolize the blood.②Administration:The rats in the puerarin group were treated with intraper- toneal injection with puerarin (dissolving in 3ml saline) after operation immediately and at 12 hours after operation. Those in the troxerutin group were treated with intraperitoneal injection with 125mg/kg troxerutin at the upper time .Those in the sham operation group and the control group were treated with 3ml saline after operation.③Observation index: The neuroetho- logy assessment ( totally 5 points ,the higher the score was ,the more severe the neurologic impairment was)was evaluated at 24h after reperfusion.The volume of cerebral infarction was assessed by TTC staining (n=5 ). The nissle body was observed by nissle staining. The expression of Bcl-2 ,Bax and TGF-β1 were tested by means of immunohistochemistry staining.Results: The neuroethology assessment of A group (0),C group (2.2±0.7),D group (2.0±0.6) and E group (2.7±0.5) at 24 h after reperfusion was lower than that of the B group (3.7±0.5), (P<0.01).
