Objective To research the relations among apolipoprotein E (apoE) genes polymorphisms, Alpha-2 macroglobulin( A2M) genes polymorphisms, Angiotensin Converting Enzyme(ACE) genes polymorphisms with Alzheimer's disease(AD) in han Chinese.Methods This research uses technology such as the polymerase chain reaction ((PCR), the restrictive fragment length polymorphism (RFLP), electrophoresis, and carrys on the related analysis to the Han Chinese in the north (114 people, 55 AD cases, 59 controls) about the apoE gene, the A2M gene, the ACE gene polymorphism .Results Between the AD cases and controls the frequency of the apoE epsilon 4 allele increased significantly in AD case ( p=0.0002,OR=5.78, 95%CI=2.30~ 14.53);between AD cases and controls the genotypes distribution of A2M have statistics significance(χ2=5.0047, P=0.0253). Two groups of allele distribution comparison also has statistics difference (χ2=4.2849, P=0.0385). I/V genotype and V allele is remarkably connected with AD (I/V genotype OR=2.85,95%CI=1.14~7.14;V allele OR=2.49, 95%CI=1.05~5.89); NO significance difference found in the genotypes and the alleles of ACE between AD cases and controls (χ2=4.5559, P=0.1025;χ2=1.0884,P=0.2968). Also after the age and the blood pressure we did not fing difference; The presence of apoE epsilon 4 allele dose not influence the ACE and A2M various genotypes and the allele distribution in AD cases and controls (p>0.05); A2M V allele dose not influence the ACE various genotypes and the allele distribution in AD case and controls (p>0.05).Conclusion apoE and A2M gene is the dangerous factors which cause late-on Alzheimer's disease .There are not connections between ACE gene and late-on Alzheimer's disease,.The A2M gene, the ACE gene and the apoE gene do not have affections each other.
