Background:Bladder cancer is the most common urologic malignancy, environmental carcinogens are considered the most important risk factor.DNA injury caused by smoking and other industrial carcinogens are repaired by DNA repair genes.Polymorphisms in DNA repair genes may be associated with alteration in DNA repair capacity and cancer susceptibility.However studies investigating the association between DNA repair gene XPD XRCC1 XRCC3 SNPs and bladder cancer yielded in consistent results.Materials and Methods:Case control studies addressing relationship between DNA repair genes XPD,XRCC1,XRCC3 and bladder cancer risk were indentified by literature search in PubMed and Embase,then relevant data were extracted from methodological eligible studies for meta-analysis.Stata 9.0 software package was used for meta analysis and Power program was used for power analysis of each study.Results:Heterogeneity and publication bias were not detected in studies of XPD.the pooled OR for XPD-751 and XPD-312 were 1.13(95%CI 0.97—1.31)and 1.24(95%CI 1.04—1.47)respectively when variant homologous genotype and wild-type allele compared.For XRCC1,heterogeneity and publication bias also not detected,the pooled OR for XRCC1-399 and XRCC1-194 were 0.95(95%CI 0.82—1.10)and 1.72(95%CI 0.73—4.04) respectively when variant homologous genotype and wild-type allele compared. Heterogeneity but no publication bias was detected in studies of XRCC3-241, the pooled OR in random effect model was 1.20(95%CI 0.89—1.61)for the TT Vs CC genotype.Conclusion:The results of this meta-analysis suggest that polymorphisms in XPD-312 increase the risk of bladder cancer.Association between XPD-751 and bladder cancer must be interpreted with caution because of it's borderline insignificant statistical result.Polymorphisms in XRCC1 and XRCC3 are not considered to be related to bladder cancer risk.
