Glycocholic acid is an important conjugated acid bile. Crude extract of GCA went through deproteinization, depigmentation, ethanol reclamation, salting-out, extraction and anhydration. Adsorption chromatography and thin-layer chromatography was respectively used in disassociation and identification of GCA. It's purity reaches to 97.3% and recovery rate is 59%.To study the anti-inflammatory effects of GCA, acute inflammation model such as ear-swelling of mice induced by xylene, skin capillary permeability of mice induced by xylene and edema of rat hind paw induced by formaldehyde and agar, chronic inflammation model such as granuloma induced by planting cotton ball in mice were adopted. The results showed that GCA could significantly inhibit acute inflammation and chronic inflammation.The contents of inflammatory media such as PGE2, NO and LTB4 were respectively measured by using UV-spectrophotometry, nitrate reductase and ELISA method to study the anti-inflammatory mechanisms of GCA. The results showed that GCA could significantly reduce the contents of PGE2 of toe swelling tissue and NO in blood serum of inflammatory rats. GCA could also reduced the contents of LTB4 in blood serum of adjuvant-induced arthritis rats. Anti-inflammtion mechanisms of GCA was related to reduce the contents of PGE2, NO and LTB4.
