PurposeDiscovery of biomarkers by use of SELDI-TOF mass spectrometry in the sera from hepatocellular carcinoma and healthy people, to search tumor markers of hepatocellular carcinoma for diagnosis.Methods25 cases of the patients with hepatocellular carcinoma without receiving any therapy , 25 cases of the patients with hepatocellular carcinoma receiving the interpose chemotherapy and 50 cases of the healthy people were tested by Weak cationic exchang(WCX2) protein chip and surface enhanced laser desorption ionization-time of flight-mass-spectrometry of Ciphergen Inc. The differentially expressed proteins were analyzed by BioMarker Wizard.Results1. At the different M/Z value range, seven proteins were obviously different in the group of the patients with hepatocellular carcinoma without receiving any therapy, the patients with hepatocellular carcinoma receiving the interpose chemotherapy and the healthy people.2. Analyzing by BioMarker Wizard software, four proteins including 6489. 48Da (P1), 6662. 34Da (P2), 8593. 75 Da (P4) and 8720. 23 Da (P5)were up-regulated in healthy controls, two including 7777. 27Da(P3) and 9250. OODa (P6) were up-regulated in the patients with hepatocellular carcinoma without receiving any therapy and one including 16200. 17 Da (P7) was up-regulated in the patients with hepatocellular carcinoma receiving the interpose chemotherapy.3. We have developed two protein profiles (7777. 27Da, 9250. OODa) that can separate patients with hepatocellular carcinoma without receiving any therapy from normal controls by use of Biomarker Pattern. It gives the much-improved sensitivity of 92% and the specificity of 100%.4. Through searching databases, we have got seven probably significant proteins related with seven different proteins among three groups.Conclusion1. Using SELDI-TOF-iWS, the method of sieving the tumor markers from HCC becomes quick and valid.2. These differential proteins could be biomarkers of hepatocellular carcinoma in the serum and drug targets for treating hepatocellular carcinoma.
