Objectives To screen a suitable protein chip from two kinds of chips (WXC2 and IMAC3). Using SELDI Protein chip technology to detect the different serum proteins spectrum of HBV infected carriers and HCC Patient, and to explore serum markers for diagnosis of HCC.Methods Serum samples from 7 cases of HCC patients and 7 cases of HBV infected carriers were collected respectively, special protein or peptide spectra was determined by SELDI-TOF-MS measurement after treating the sample onto two kinds of protein chips (WXC2 and IMAC3) for each case. According to the protein profiling results, screening suitable protein chip. Surface enhanced laser desorption /ionization time-of-flight mass Spectrometry technology was used to analyze sera including 44 HCC Patients,55 HBV infected carriers on WCX2 protein chip.The datas were analyzed by Biomarker Wizard and Biomarker Patterns Software 3.2(BPS3.2),and then a diagnosis model was setup.Results Analysis of serum protein fingerprints showed WCX2 chip combined more proteins than IMAC3 chip. Comparison of the group of hepatocrllular carcinoma and and the group of HBV infected carrier: analysis two groups of 99 samples and get 190 proteins peaks, among them 117protein peaks show the difference obviously in two groups(p<0.05). Compare with the HBV infected carrier group, 39 proteins were up-regulated expression and 78 proteins were down-regulated expression. Make use of 117 proteins of those differenced proteins, we can get a model. Sensitivity of this model is 95.5%(42/44), specificity is 92.7% (51/55) on diagnosis of HCC.Conclusion WCX2 chip can detect more significant difference protein peaks which have distinguished the HCC patients from the HBV infected carriers compared to IMAC3 chips and preferably apply to the research of serum protein fingerprints of the hepatocrllular carcinoma. The high specificity and sensitivity in this study provide support of the potential for SELDI protein chip technology combine with bioinformatics technology to improve the early diagnosis of HCC,the prospect of clinical application is bright.
