Objective: To determine frequencies distribution of peroxisome proliferator activated receptor γ 2(PPAR γ 2) gene Pro12Ala polymorphism genotype and allelein Han and Korean population of Yanbian area in China, and relationship with type 2 diabetes mellitus. Methods: Polymerase chain reaction-restriction fragment length polymorphism was used to test a coding variant Pro 12Ala of PPAR gene in 49 Han people and 51 Korean people with normal glucose tolerant (NGT) ,and 52 Han patients and 53 Korean patients with type 2 diabetes mellitus (DM) .Case-contrast analysis was used to study the association between Pro12Ala polymorphism and type 2 DM. Results: (1) The A allele frequencies of NGT subjects were 5.10% for Han, 5.88% for Korean, and no significant difference was found between this two groups( x~2=0.059, P > 0.05); (2) The A allele frequencies of type 2 DM patients were 2.89% for Han, 3.77% for Korean. Allele frequency distribution of each ethnic was compared between DM group and NGT group. There were no significant differences( x~2=0-20,P>0.05; Han; x~2=0149, P>0.05 for Korean); (3) There was no association between PPAR 7 2 gene Prol2Ala polymorphism and the level of BMI, FPG, TC, TG, HDL-C, LDL-C in Han type 2 DM group and Korean type 2 DM group.Conclusion: There was no difference in frequencies distribution of PPAR gene Pro 12Ala polymorphism between Han and Korean population of Yanbian area in China. Their allele frequencies were similar to Japanese, however, there were significant differences when compared with European White. The Prol2Ala polymorphism was not associated with type 2 DM in Han or Korean population ofYanbian area in China;PPAR y 2 gene Prol2Ala polymorphism has significant ethnic differences.
