The disease incidence increasing with the the ageing becamesevere,multiple factor have significant relationship with the happenand development of AS,the increasing of Ang II and the decreasingof NO is early marker of disproportion of endotheliocyte.they mayincrease the permeability of VEC and intimal membrane,add theintension of vascular;LDL especially OX-LDL may stimulate theproliferation of smooth muscle,MMPs can remodule thematrix ,VEGF can promote the development and the instability ofthe plaque,VCAM-1 can mediate the adhesion of cell-cell andcell-matrix.out factor can advance the development of AS throughsuch biological effect.what's more ,such effect may be transferredthrough NF-γB pathway.traditional Chinese medicine is a newpathway to cure AS,very many kinds of medicines can suppress thehappen and develop through above-mentioned biological effect.butthe the study on the effect of panaxoside to cure AS is few.we observe the effect of anti-anthrosclorosis of panaxosidethrough detecting the blood fat , Ang II,NO,NOS,MMP-9,VEGF,VCAM-1,NF-γB and the change of proliferation of arteryintimal membrane before and after cured by panaxoside. We madethe mode of artherosclerosis of rat through feed artherosclerosisfeeds add the sacculus proprius damage methods,dividing such ratsinto two groups according to the different supplying panaxosidemethods,at the same times,the normal control group and shamoperation group were made.the TC,TG,LDL were detected inBackmaanLX20 autobiochemistry analysator.detecting theconcentration of NO,NOS in peripherial blood through NO,NOS kit.Ang II was detected by radio immunoassay method,morever,wedetected the expression of MMP-9,VEGF,VCAM-1,NF-γB intissue through RT-PCR,westernblot,immunohistochemical in situapproach.observe the change of intimal and tunica media with elasticfibers staining and calculate the ratio of intima and tunica media.splitthe aorta along axis of ordinate and stain it with HE method,changethe image analystor into number analystor,calculate the the ratio ofthe area of intima plaque and the area of tunica media.The detection of each factor and blood lipoid demonstrate theactivity of NO,NOS decreased ,the TG,TC,LDL,AngⅡ,VCAM-1,VEGF,MMP-9,NF-γB increased, I/M,ratio of plaquearea,proliferation index increase compared normal control groupand sham operation group.panaxoside can modulate the change ofobove-mentioned factor according to opposite direction.the cureeffect of drug prophylaxis group is better than the drug treatment.theTG,TC,LDL,AngⅡ,VCAM-1,VEGF,MMP-9,NF-γB,NO,NOS,I/M,ratio of plaque area show siginificant defferencebetween such two groups after cured by panaxiside.panaxoside can cure and protect the rat from AS and the curedeffect of drug prophylaxis group is better than drug treatment group.All of a word,The activity of NO,NOS decreased;the TG,TC , LDL , Ang Ⅱ, VCAM-1 , VEGF , MMP-9 increase inartherosclerosis module.such factors play a crucial role in thehappeness and development of early artherosclerosis.panaoxside candeteriorate the change of above-mentioned factors.the change maybe mediated through NF-γB pathway.
