Objective:To explore the effects of sinomenine on athererosis model of rabbits the level of lipid metabolism in carotid artery pathology morphology;To explore the impact of the epoxide COX-2expression in the organization of sinomenine the rabbit carotid artery atherosclerosis;To expand of sinomenine the clinical application of space to provide the exactly experimental basis.Methods:Fourty-eight rabbits were chosen and randomly divided into six groups;Detected the rabbit’s weight in different times;Taken the neck artery and did haematine and eosin staining;Detected the inflammation-related indicators and lipid metabolism-related indicators in serum with enzyme-linked immunosorbentassay;Detected the expression of COX-2mRNA in neck artery by RT-PCR;Figured and analyzed the impact of sinomenine to the indicators.Results:1、SIN can make rabbits’weight losing.and with the increasing of the dose and giving time,weight losing is more obvious. SIN has a significantly reduce atherosclerosis model in rabbit serum of TG、TC、LDL-C and HDL-C levels. And with the increasing of dose,the greater the impact lipid metabolism.2、SIN has a significant of atherosclerosis model of rabbit carotid arterytissue of C0X-2mRNA expression, and with the increasing of COX-2mRNA expression,the dose was negatively correlated.3、SIN anti rabbit carotid atherosclerosis,a significant protective effect of rabbit carotid intima,and carotid intima injury severity and the dose was negatively correlated.Discussion:SIN has the obviously role of anti-rabbit carotid artery artherosclerosis; Sinomenine can notablely inhibition of carotid intimal injury and the generating of inflammation-related media;it can obviously adjust abnormal of lipid metabolism-related indicators and the expressing of COX-2mRNA in artherosclerosis tissue.With the increasing of sinomenine’s doses,the protection of the carotid intima is more obviously,and the greater the inflammation associated media,the level of lipid metabolism,as well as the expressing of COX-2mRNA in carotid artery tissue.Therefore,we conclude that:sinomenine can inhibit the expression of COX-2mRNA in rabbit neck artery atherosclerotic tissue,so as to achieve the formation of the inhibition of atherosclerosis.
