Objective: To investigate the expression of BDNF in the region of cerebral cortex and basal ganglion and the possible meaning of the expression in the Guinea Pig' s kernicterus model. Methods: The guinea pig model was made by the way of celiac injection of bilirubin. The experimental animals were randomly divided into five groups: control group, 100mg/kg groups (4h and 8h), 200 mg/kg groups (4h and 8h). Every group include 10 animals, 4~6 days old. After the treatments, the animals were sacrificed, perfused and fixed in corresponding time points and then 20μm frost-slices were made and treated with immunohisto- chemistry. The positive-expression neurons were counted under micro- scope and the data were processed with one way ANOVA and least significant difference (LSD) method. Results: (l).In control group, BDNF expressed in both the cortex and basal ganglion .Expression BDNF in the cortex region is more than the basal ganglion .(2).BDNF expression in cortex: 100 mg/kg group(4h) and 200 mg/kg group(4h) decreased compared with the control group but T24h group is no significance between them (P>0. 05). 100 mg/kg group(8h)and 200 mg/kg group(8h) increased compared with the control group but T18h there is no significance between them. (P>0. 05). (3). BDNF expression in basal ganglion: 100 mg/kg group(4h)and 200 mg/kg group(4h) decreased compared with the control group and there is significance between the 100 mg/kg group(4h) and control group while there is not between 200 mg/kg group(4h) and the control group. 100mg/kg group(8h) and 100 mg/kg group(8h) increased and the statistical significance is opposite with the former. (4) BDNF expression in the different groups: Between 200 mg/kg group(4h) and 200 mg/kg group(8h), the cortex region is increased compared with the basal node region and there is significance between them (p<0. 05) and it is the same result in 100 mg/kg groups(4h and 8h). Conclusion: (l).Both the cortex and the basal node region can express BDNF and it may be related with the survival of the neurons. (2).The expression of BDNF in the region of cerebrum cortex is stronger than the ganglion basilare. (3). In the initiated period of injury, the expression of BDNF is decreased and with the injury delayed and aggravated , the expression of BDNF increased, indicating that BDNF may has dual functions in kernicterus.
