Objective:1. Founding of profiling of comparative proteomics of focal cerebral ischemia on model rat in basal ganglion, to find the proteins related with cerebral ischemia, to explore pathologic mechanism of it.2. By acupuncture method of "Xing Nao Kai Qiao" originated by academician Shi xue-min, to study acupuncture effect on proteins expression of focal cerebral ischemia model rat.Methods:The Wistar rats were random divided into three groups: normal group, model guoup and acupuncture group. Each group was divided into two inferior groups: 6h group and 24h group. Using nylon monofilament to make middle cerebral artery occlusion (MCAO) model, to give a mark of neural function defect on the rats after they waking, the marks reach to 3 is the level to be success model. Selecting Nei Guan, Ren Zhong points to puncture on acupuncture group, electric needle parameter as: frequency:2HZ, electric current: 3mA, time: lmin. Each group was to guillotine and get the brain in defined times. Then dissecting ischemic basal ganglion and extracting total proteins of basal ganglion to perform 2-DE. The gel was dyed with Coomassie brillint blue R-350. The images were analyzed by software of Image master 2D V3.01 Elite. Selecting differentially expressed proteins to march enzymolysis in gel, detect peptide mass fingerprint by MALDT-TOF-MS, then research data base of Swiss-port to identify the proteins. Results:1. Compared with model group and control group, there were 17 differentially expressed proteins. Ubiquinol-cytochrome c reductase iron-sulfur subunit expressed only in model groups. In model 6h and 24h groups 5 increased proteins were: Glyceraldehyde-3-phosphate-dehydrogenase, Ubiquitin carboxyl-terminal hydrolase isozyme L1, Glutathione S-transferase P, Anti-oxidant protein 2, Tissue-type...
