| [Objective]To investigate the relationship among the levels of serum thrombopoietin (TPO) , the mRNA levels of thrombopoietin receptor (c-mpl) on platelet , peripheral blood platelet ( PLT ) counts , megakaryocyte counts in patients with thrombocytopenia and it's clinical significance.[Methods] The levels of serum TPO, the mRNA levels of c-mpl on platelets, PLT and megakaryocyte counts were detected in 50 patients with thrombocytopenia and 20 healthy volunteers(normal controls) by using sandwich-ELISA, semi-quantitative RT-PCR, automatic hemocytoanalyser and handmade count. [Results] The levels of serum TPO and the mRNA levels of c-mpl on platelets in patients with idiopathic thrombocytopenic purpura (ITP) were low lightly compared with those in normal controls(p>0.05); The levels of serum TPO and the mRNA levels of c-mpl on platelets in patients with secondary thrombocytopenia were higher than those in normal controls (p<0.05); In patients with ITP , the levels of serum TPO and the mRNA levels of c-mpl on platelets were not correlate with PLT , megakaryocyte counts (p>0.05); the levels of serum TPO were positive correlate with the mRNA levels of c-mpl on platelets(r = 0.63,P<0.05). In patients with secondary thrombocytopenia, the levels of serum TPO were negative correlate with PLT and megakaryocyte counts (r = -0.68,P<0.05; r = -0.58,P<0.05); the mRNA levels of c-mpl on platelets were negative correlate with PLT and megakaryocyte counts(r --0.65,P<0.05 ; r = -0.55,P<0.05) . In patients with ITP and secondary thrombocytopenia, the levels of serum TPO were positive correlate with the levels ofc-mpl mRNA on platelets (r - 0.63,P<0.05; r = 0.70,P<0.05).[Conclusion] Measurements of serum TPO levels and c-mpl mRNA levels onplatelets have certain diagnostic value in thrombocytopenia. The levels of serum TPOand mRNA levels of c-mpl on platelets were regulated by PLT and megakaryocytecounts. |
