The topic utilized modern technology of phytochemistry and apparatus analyze in Studies on Rhizoma Dioscoreae Nipponicae and Radix Angelicae Pubescentis. The paper includes 2 parts:The first part: Studies on Radix Angelicae Pubescentis. Summarized the evolution of Radix Angelicae Pubescentis and fingerprint about traditional Chinese medicine.To establish a method for the determination of Osthole in Radix Angelicae Pubescentis by HPLC. The chromatographic condition was follows: 4.6mm×250mm, peak with ODS (particle size 10μm); mobile phase :acetonitrile—water(6:4) ; flow rate :1.0mL.min~-1; detection wavelength: 322nm; Osthole showed good linearity(r =0.9999) in the range of 10~150μg.mL~-1;The average recovery: 99.5%(n=9). This method is successfully usedfor the assay of Radix Angelicae Pubescentis.According to the technical demand of fingerprint about traditional Chinese medicine ,We formulated the fingerprint's examination standard of Radix Angelicae Pubescentis.There are 10 characteristic peak in Radix Angelicae Pubescentis .It was 80 minutes that HPLC collected the chromotogram signal. Similarity of each batch of injection was from 0.85 to 0.93 except 1 sample. As the result the method of fingerprint was accurate, repeatable and steady.The second part: Studies on Rhizoma Dioscoreae Nipponicae. Include the evolution ,separation of Rhizoma Dioscoreae, Nipponicae.To develop a new quantitative analysis method for the determination of dioscin in Rhizoma Dioscoreae Nipponicae and di'aoxinxuekang Capsules by HPLC-ELSD.Rhizoma Dioscoreae Nipponicae: 10kg, dioscin:39.2g. The separation of dioscin :0.39%.Developed a new quantitative analysis method for the determination of dioscin in Rhizoma Dioscoreae Nipponicae and di'aoxinxuekang Capsules by HPLC-ELSD. Chromatographic: The peak of dioscin has a retain time at 9.9 Min. Thechromatographic condition was follows: chromatographic column : 4.6mmx250mm, peak with ODS (particle size lOum); mobile phase: acetonitrile and water (55:45); mobile phase flow rate^.OmL'min^column temperature was ambient temperature; clear air pressure:5*105Pa and drift tube temperature:90.3°C. Dioscin in Rhizoma Dioscoreae Nipponicae was well-separated.The linear calibration curve was obtained in the concentration rang of 80~600ug-mL'' (r = 0.9998) . The average recovery of dioscin was 99.88%, RSD =1.25% (n = 9) . Dioscin in di'ao xinxuekang Capsules was well-separated.The linear calibration curve was obtained in the concentration rang of ^(MSOOug-mL"1 (r = 0.9999) . This method is simple, accurate and reproducible.lt is also proposed to be used for quality control of Rhizoma Dioscoreae Nipponicae and di'ao xinxuekang Capsules.
