[Abstract]Objective: Lymphatic metastasis is the main metastasispathway in colorectal carcinoma ,but the concrete mechanism is notclear still.1 The experiment is to use the immunohistochemical doublestaining to study the distribution and morphological character ofmicrolymphtics in colorectal carcinoma ,peritumoral tissues , normalcolorectal tissues ;2 To analysis the ralationship of lymphaticmicrovessel density ,maximal profile average area and averageperimeter in colorectal cancer of anisohistology typing ,in orderto discuss the metastasis pathway of tumor cells deeper and thebiological behavior of colorectal cancer .Method:Randon collection of the preoperative untherapeuticanisohistology typing colorectal cancer : DukesA phase 20 pou,DukesBphase 20 pou,DukesC phase 20 pou,DukesD phase 20 pou and normalcolorectal tissues 20 pou paraffin masses ,distribute to fivegroups ,all the paraffin masses process routine section and use theimmunohistochemical double staining,the proceed ofimmunohistochemical double staining is accoding to the reagentinstruction of American ZYMED company ,using UEA-1 and monoclonalantitype IV collagen anti-body to the tissue sections ,then observethe lymphatic capillaries ,blood capillaries and use stereologicalmethod to put the mono-network test system of cubic-network systeminto the light microscope and take the sections into the lightmicroscope ,so as to observe and assay lymphatic microvesseldensity,maximal profile average area and average perimeter , thenexert calibrator to calibrate ,the regions of assay are selected theperitumoral tissues and normal colorectal tissues Results: 1 only microlymphtics endothelial cells are stained bluecolour ,but blood capillary are stained double-deck pigmenting,bloodcapillary endothelial cells show blue and brown colour , bloodcapillary basement membranes show red colour ,the difference is veryclear .2 In the tissue sections , there is not transparent lymphaticsin colorectal cancer ,but in the peritumor matrix there are muchlymohatics . 3 There was significant difference between differentphase colorectal carcinoma peritumoral tissues lymphaticmicrovessel density(average value: 20.9 士8.06) and normalcolorectal tissues(average value: 10.1 士2.34) ( P<0.05 ) ,lymphatic microvessel density was significantly higher in lymph nodemetastasis grope(average value: 27.45 士5.89) than those in nometastasis grope(average value: 14.325 士2.89) (P<0.05) ,butbetween lymph node metastasis grope or no metastasis grope , therewas no significant difference ;4 There was significant differencebetween different phase colorectal carcinoma peritumoral tissuesmaximal profile average area(average value: 1476.737 士557.94) andnormal colorectal tissues(average value: 715.902 士165.37)(P<0.05) , maximal profile average area was significantly higherin lymph node metastasis grope(average value: 1938.38 士416.37) thanthose in no metastasis grope(average value: 1015.09 士141.63)(P<0.05) ,but between lymph node metastasis grope or no metastasisgrope , there was no significant difference ;5 Different phasecolorectal carcinoma peritumoral tissues average perimeter(averagevalue: 45.587 士6.18) are significantly higher than those in normalcolorectal tissues(average value: 35.659士5.48)(P<0.05),but therewas no significant difference between different phase colorectalcarcinoma peritumoral tissues(P>0.05). Conclusion:1 Immunohistochemical double staining method couldaccurately differentiate microlymphatics from blood capillary andis a objective method in studying the distribution of microlymphaticsand the quantitive change of microlymphatics in colorectal carcinoma2 There may existence lymphangiogenesis by the colorectal carcinomaperitumoral tissues from high differentiation to poordifferentiation ,it may say that lymphangiogenesis is an importantrole in metastasis and relapse in colorectal carcinoma .3 Use targetand selection to block lymphangiogenesis may ameliorate the prognosiof colorectal carcinoma .
