| Understanding the structure and function of the brain is the most challenging frontier scientific problem in the world.The cerebrovascular network is the most important energy supply structure for the brain.The study of brain network structure provides an important basis for the analysis of brain functional imaging information,changes in brain development and functional realization,and cerebrovascular disease.With the rapid development of dyeing technology and imaging technology,the whole brain fine blood vessel network data set has been obtained,which provides the basis for the study of three-dimensional reconstruction and quantitative analysis of multi-scale vascular network.However,limited by the reconstruction method and computing resources,at present,the study of capillaries in the fine blood vessel network is still limited to the cortex,cerebellum and other regions.There is still a demand for three-dimensional morphological reconstruction and quantitative analysis of the capillary network in subcortical structure.The high-resolution mouse whole brain three-dimensional image data set was obtained by the Micro-optical Sectioning Tomography combining with the improved whole brain Nissl staining technique contained cell and blood vessel construction information.Based on the data set,a set of standardized brain area levels of capillary network three-dimensional reconstruction and quantitative analysis process was established.The process consists of three parts:(1)three-dimensional reconstruction of fine blood vessel network in mouse cortex and subcortical structure;(2)based on vascular diameter,the vascular mean diameter,vascular fractional volume and vascular normalized length of different capillary networks were calculated;(3)multilevel quantitative analysis of fine blood vessel network in mouse brain was carried out based on the results of parameter calculation.In this paper,the results of three-dimensional reconstruction of fine blood vessel network in several brain regions of different age groups were obtained.The distribution of capillary network among different brain regions in adult mice,the distribution of capillary network in the left and right hemispheres and the trend of the changes of the brain blood vessel network in the developing stage were studied by quantitative analysis.The work of this paper provides a basic research data for the whole brain fine blood vessel network research. |
