Background and objectives: Bronchial asthma (abbreviating asthma) is a chronic inflammation of airway, which is induced by a series of inflammatory cells and structure cells , such as mast cells , eosinophils, T lymphocytes and so on. Stem cell factor (SCF) is a main cytokine that activiates mast cells and eosinophils Therefore, SCF may take part in pathophysiology process of asthma. Especially, with more and more importance has been attached to SCF, such as in hemotoposis system and the success of synthesis of human recombine SCF, SCFASON and the antibody of the SCF and have been applied in some area. It may provide new target and method for the treatment and diagnosis of asthma to reveal the role of SCF in pathophysiology process of asthma, especially the different roles of the two forms SCF.There are two differert forms SCF: soluble stem cell factor( sSCF) and membranous stem cell factor (mSCF )because different mRNA cutting site. These two types SCF have diverse roles, sometimes even opposite ones. The role of sSCF has not been studied , especially there is no study on variances and significances of sSCF in the patient with asthma all over the world . In the experiment, we use enzyme-linked immunosorbent assay(ELISA) to measure serum sSCF andimmunoglobulin E(IgE) levels and count the number of EOS directly in 92 children with asthma, 33 normal children to invest variances and significances of serum sSCF in children with asthma. In the meantime, we also measured the variances of sSCF, IgE and EOS after using dexamethasone to reveal the role of sSCF in pathophysiology process of asthma for the more. This may provide new target and method for the treatment and diagnosis of asthma.Materials and Methods: Using ELISA to measure serum sSCF and IgE levels and count the number of EOS directly of 60 children with asthma in acute phase, 32 children with asthma in remission phase, 33 normal children, age, sex and so on are not significantly analyzed by statistics(P>0.05).And the variances of sSCF, IgE and EOS after using dexamethasone is measured .Using SPSS 11.0 statistical software , mean and standand( x ± s ) indicate results, every group parameters conform to normal distribution, sample averages of every group are analyzed after analysis of even variance, after that using t and F test, using linear regression, linear correlation analyse, P value less than 0.05 indicate statistical significance.Results: 1.Serum sSCF concentration (2.9134 ± 0.7596)pg/L is significantly lower in acute phase of asthma than in period of remission (3.5004 ± 0.5985)pg/L (P<0.01), whereas period of remission and control group(3.7163 ± 0.5775)pg/L are significantly different(P<0.05); Serum IgE concentration(259.2±61.8437)U/L, the count of EOS (0.3405±0.1176) ( × 10~9) in acute phase of asthma are significantly higher than in period of remission IgE( 190.1 ±64.3321)U/L, EOS(0.2336±0.1342) ( × 10~9) (P<0.01) .Furthermore, they are significantly higher than control group IgE (102.2±1.8956) U/L,EOS(0.1091± 0.1510) (× 10~9) (P<0.01), view in table 1. 2. Serum sSCF concentration (2.6890± 0.6874) pg/L in medium, serious asthma are significantly lower than light asthma (3.0345 ± 0.6489) pg/L, (P<0.05) ; Serum IgE(279.3±71.3124) (u/L) concentration and the count of EOS (0.3858 ± 0.1533) ( × 10~9 ) in medium, serious asthma are significantly higher than IgE (239.2±64.8216) (u/L), EOS (0.3045±0.1423) ( × 10~9) (P<0.01) in light asthma, view in table 2.3.Serum sSCF concentration (3.1904 ± 0.7089) pg/L after using dexamethasone are significantly higher than the one before treatment(2.9134 ± 0.7596) pg/L, Serum IgE concentration (210.3±64.9087) (u/L) and the count of EOS (0.2532±0.1312) ( X 109) after using dexamethasone are significantly lower than IgE(259.2±61.8437) pg/L and EOS (0.3405±0.1176) (X109) (F<0.01) before treatment, view in table 3. 4.There is negative relation among serum sSCF and IgE concentration and the count of EOS in the acute and remission phase of asthma. There is positive relation with the count of EOS and IgE concentration in the acute and remission phase of asthma ,view in table 4.Conclusions:1. sSCF may restrain the process of asthma.2 Measuring the serum sSCF concentration is useful for monitoring development, remission diagnosis and theraphy of asthma.3 sSCF may be a kind of the mechanism of dexamethasone in asthma treatmeat.
