Objective: This study was designed to evaluate the influence of different laser photostimulation time on morphometric and biochemical analyses in experimentally tenotomized and repaired rabbit Achilles tendons and to deserve the best photostimulation time. Methods: A total of 24 male New Zealand rabbits were used. Following tenotomy and repair. The experimental animals were divided among three groups: laser therapy 10 minute, 20 minute, 30 minute. The experimental animals were treated with He-Ne laser daily at l.0Jcm~2 for 14 days. Control animals were sham trented with the laser head. We used HE stain and Van Geison stain methoed to observe morphometiic of tendons. The Achilles tendons were excised, Processed and analyzed by measuring hydroxyproline. The type of collagen and ratio was subsequently analyzed by SDS-PAGE and CS-930 light density scan instrument. Results: Histological evaluation showed that laser irradiation enhanced cell proliferation, cellular content, granulation tissue formation, and collagen deposition in laser-treated tendons as compared to the control group. Biochemical analyses of the tendons revealed a increase in collagen with laser photostimulation indicating a more rapid healing process in 20 minutes treatedgroup compared to controls. The results indicated that type I and type III collagen levels were markedly increased at 28 days of healing tendons with He-Ne laser and the ratio collagen III to I increased in control group response to experimental groups. Conclusion: The results suggest that laser photostimulation promotes the tendons repair process. The proportion of type III collagen was more rapidly than in the control group. The best laser photostimulation time is 20 minutes.
