Objective:To establish rat renal transplantation of acute rejection models,it was discussed that the mechanism of Mat resists acute rejection of renal allograft and synergistic effects with cyclosporine A (CsA). The immunosuppressive efficacy of Mat was discussed in order to improving relating theory-basis for further developing and the machanism of synergistic effects with low-dose CsA.Methods:Establishing rat renal allograft models with acute rejection (Donor:inbored SD rat Recipient: inbored Wistar rat).48 recipients were divided in to 4 groups at random.Group NS was treated with NS(1ml/kg.d ip);Group CsA was treated with Cyclosporine A(2.5mg/kg.d ip);Group Mat was treated with Matrine (10mg/kg.d ip);Group Mat + CsA treated with Mat (10mg/kg.d ip) combination with CsA (2.5mg/kg.d ip).Establising isograft models (Wistar-Wistar) ,12 recipients as a group was used as a control and were treated with NS (1ml/kg.d ip).The allograft survival time and 24hr-urinary volume were observed.Allografts of 6 recipients from each group were killed at random at day 7 after renal transplantation ,vein blood was received to monitor levels of Cr and Bun ,levels of IL-10 and CD44 in vein blood were monitored by enzyme-linked immunosorbent assay (ELISA).The transplanted kidneys were used to observe the patholoyical changes. The survival time and 24hr-urinary volume of other recipients were observed .All these indicator were used to analyse the efficacy...
