The Effect Of Matrine Treatment On The Expression Levels Of IL-2 And TNF-α In Vein Blood Of Rat Renal Transplantation Models

Object i ves:To evaluate the immunosuppressive efficacy and outcome of matrine inacute rejection after rat renal allograft,to evaluate the synergistic effect in combination with Cyclosprin A.This research is based on the rat renal allograft models. The choosed indicators are the levels of IL-2 and TNF- α in vein blood,This study aims at:the use of Mat can reduce the dosage of CsA and the expenditure of patients;providing theory foundation to clinical appliance of Mat;a novel and safe scheme can be offered to clinical organ transplantation.Methods: We established the acute rejection animal models with inbred SD beingdonors and inbred Wistar being recipients, in which allograft acute rejection occurred at 7 days posttransplantation. The recipients were divided into 7 groups at random: Group N.S, as experiment control were treated not with immunosuppressants, only with 0.9%Sodium Chloride (N.S,1ml.kg^-1.d^-1,i.p); Group Mat,treated with Mat (10mg.kg'M'^i.p); Group CsA,treated with CsA(2.5mg.kg^-1d^-1,i.p); Group Mat+CsA,treated with Mat (10mg.kg^-1d^-1_,i.p in combination with CsA(2.5mg. kg^-1M^-1,i.p); Another 12 isograft recipients with inbred Wistar being donors and recipients as group V,and group VI with 12 inbred Wistar as surgical controls ,and group VII with 12 inbred Wistar as normal controls were not treated with immunosuppressants,only with N.S(1ml.kg^-1.d^-1,i.p) respectively.The allografts survival time and 24hr-urinary volume within four weeks were observed; BUN and Cr in blood were measured; Halves of grafts or kidneys from each group were harvested for pathomorphism analysis respectively at days 7 postoperation. The data were analysed by using the one-way ANOVA and/or Dunnett's T3 respectively.All these indicators were used to analyze the efficacy of Mat,CsA,or Mat in combination with CsA.Results: Control recipients survived over 100 days.Recipients of Group N.S died Less...
