Objective : TO investigate a C/T polymorphism in the 5'-untranslated region (UTR) of the CD40 gene and it's relationship with Graves' disease of hepatic injury in Tianjin , China. Methods: The CD40 5'UTR SNP (C-T,position-1) was amplified by polymerase chain reaction (PCR) and genotyped using the restriction enzyme NcoI . The role of the 5'UTR in the susceptibility for GD of hepatic injury among the 60 GD patients with hepatic injury and 60 GD patients and 106 normal subjects was investigated.Results. 1. Case control association analysis of the CD40 C/T-1 SNP in 60 GD patients without hepatic injury, 60 GD patients with hepatic injury and 106 normal subjects showed significant difference. But there were no differences between GD patients with hepatic injury and GD patients without hepatic injury; 2.CD40 C/T.] SNP did not show difference among normal subject, GD patients with ophthalmopathy and GD without ophthalmopathy, 3. CD40 C/T-1 SNP did not show difference among normal subjects , GD patients with goitre and GD patients without goitre ; 4. CD40 5'UTR genotype showed difference between GD patients with leukopenia and GD patients without leupopenia,CD40 5'UTR allele and phenotype did not show difference between two groups. Conclusion: Our findings suggest that 1.CD40 gene 5'UTR C/T-1 SNP may...
