Background : Insulin Resistance is specified that the biologic effects of some insulin combined with its idiosyncrasy acceptor are lower than normal level. Insulin resistance induce to a cluster of metabolic abnormalities ,including obesity with distribution of the fat in the central part of the body , hypertension , impaired glucose tolerance , dyslipidaemia , fibrinolytic abnormity. Some studies showed that IR is correlated with cardiovascular diseases, furthermore fibrinolytic system activity abnormity of coronary heart disease is put more attentions. Tissue-type plasminogen activator (t-PA)and plasminogen activator inhibitor 1 (PAI-1) are the key to adjust fibrinolytic system .Thus, it is helpful for exploring the mechanism that IR induce to coronary heart disease, to learn on the level of PAI-land t-PA in insulin resistance .Objective : Making insulin resistance modal of rats by high fat feeding , we test the level of plasma PAI-1 and t-PA , in order to find the effect from insulin resistance to fibrinolytic system. To explore the effect of rosiglitazon maleate ,Jinqi tablet on improve insulin resistance and PAI-1, t-PA activity.Methods : Wister rats(6 weeks old) were divided into two groups in random taking either normal chow(NC,12% as fat , n=9),or isocaloric high-fat diet (HF, 59% of calories as fat , n=9). FPG and FINS were tested before feeding, FPG, FINS, PP2hPG, PP2hINS were tested after eight weeks, ISI was calculated , so insulin resistant modal was made . Then , HF group was divided into A,B,C three groups in random .A group is modal group (high fat feeding ,n=9), B group is Avandia group(high fat feeding Mvandia, 3mg/Kg/d, n=9), C group is Jinqi group(high fat feeding + Jinqi tablet, 2.lg/Kg/d,n=9) and D group is MC. Test FPG, FINS, PP2hPG, PP2hINS of four groups after feeding fourteen weeks , than killed them .Test visceral fat mass and plasma TG, TC, FFA of four groups .The activity of PAI-1 and t-PA were assayed by chromogenic substrate method.Results ".There was no significantly difference in the body weighbetween two groups before feeding. The body weigh of high fat (HF) group was significantly higher than normal chow(NC) group after feeding eight to fourteen weeks , P<0. 05. After feeding fourteen weeks , A group was significantly higher than D group in visceral fat mass/body weight, P<0. 01. There was no difference in FPG and FINS between HF and NC before feeding ,P > 0.05. Feeding eight to fourteen weeks , HF group was significantly higher than NC in FPG and PP2hPG , P<0. 001, so was in FINS and PP2hINS, P<0. 05. B and C groups were significantly lower than A group in FINS and PP2hPG, P<0.001. After feeding eight to fourteen weeks , HF group was significant lower than NC group in insulin sensitivity index(ISI), P<0. 001. B and C two medicine groups were significant higher than A group in ISI, P<0. 01. After feeding fourteen weeks A group was significant lower than D group in t-PA , P<0. 001, B and C groups were significant higher than A group , P<0. 01. There was no difference between B and C group .A group was significant higher than D group in PAI-1, P<0. 001, B and C group were lower than A group, P<0. 01.Couclusions:1.
