Objects: The study was designed to examine the influence of penehyclidime hydrochloride and atropine on the circulation of rats with omethoate poisoning, for a rather therapy to the patients with organophosphate poisoning. Methods: The number of the Wistar rats was totally 96,among them 24 for A experiment and 72 for B experiment. The rats was divided randomly into four groups in average: the brine matched control group; the omethoate intoxication matched control group; the omethoate intoxication and atropine group; the omethoate intoxication and penehyclidime hydrochloride group.The number of rats in each group was 6 in A experiment, 18 in B experiment. And then each group in B experiment was divided evenly into 24 hours team, 72 hours team and 7 days team with random, and the number of rats in each team was 6. The poisoned model was made by omethoate (100mg/kg) subcutaneous injection. The heart rate (HR),blood pressure (BP) and Electrocardiogram (ECG) of the anesthetized rats were monitored by BL-410 experiment system in A experiment before giving medicine and 15 minutes,30minutes,1 hour and 2 hours after giving medicine. The blood samples were taken for detection of LDH, CK and CHE two hours after giving medicine. In B experiment, the appear and disappear time of organophosphate poisoning symptom was observed in conscious freely moving rats, and the heart specimen was taken for histological confirmation at 24 hours, 72 hours and 7 days after giving medicine. Results: The rats in the brine matched control group moved freely and had no symptom. The saliva, fasciculation, dyspnea and miosis signs of rats appeard early and the performance was heavy in the omethoate intoxication matched control group, all of them in B experiment died quickly. The saliva and miosis signs of rats weren't observed in the omethoate intoxication and atropine group, the fasciculation and dyspnea signs were alleviated gradually two or three hours later, and one of them in B experiment died about 20 hours later. The saliva and miosis signs of rats weren't notable in the omethoate intoxication and penehyclidime hydrochloride group either, the dyspnea and fasciculation signs of rats were lighter and the active recovering time of rats were earlier than the rats in the omethoate intoxication and atropine group. The heart rate (HR) of rats descended slightly in the brine matched control group, and dropped rapidly all the times in the omethoate intoxication matched control group. The HR of rats had a sharp increase and then decrease quickly to a stable level in the omethoate intoxication and atropine group, but descended smoothlyto a balanced state in the omethoate intoxication and penehyclidime hydrochloride group. The changes of R-R period were the main electrocardiographical abnormities of experimental rats, conducting defects and other arrhythmias. The blood pressure (BP) of rats had no prominent change in the brine matched control group, while showed a marked increase and then declined slowly to a normal level in the omethoate intoxication matched control group and in the omethoate intoxication and atropine group, but only enhanced slightly in the omethoate intoxication and penehyclidime hydrochloride group. The activity of CK,LDH of rats in the omethoate intoxication matched control group was the highest in these groups, and among them the activity of CK in the omethoate intoxication and atropine group was higher than that in the omethoate intoxication and penehyclidime hydrochloride group. The activity of CHE was lower markedly in the omethoate intoxication matched control group and in the omethoate intoxication and atropine group than that in the brine matched control group, while the activity of CHE decreased slightly in the omethoate intoxication and penehyclidime hydrochloride group and showed no marked difference with that in the brine matched control group.
