Acute organophosphorus pesticides poisoning (AOPP) is a global problem that imperils human health. at present, the death rate is reported 4-30% in different countrys and regions. AOPP can arouse a series of syndrome with myasthenia as cardinal symptom, being reported and amed after to intermediate myasthenia syndrome (IMS) at first by scholar Senanayake of Sri Lanka in 1987. Respiratory muscle paralysis (RMP) is the crisis that has most the clinic salvage meaning among IMS. After the occurrence RMP, The curative effect of atropine and pralidoxime chloride is not good. Therefore, it has all-important significance to study the cause of RMP conduced by AOPP and look for the reasonable valid prevention and cure medicine. There are two major sorts of counterpoison to cure AOPP: atropine and pralidoxime chloride. It is the consensus of all clinical doctors to use the full quantity of atropine again and again, attain the "atropinfy"quickly, in the AOPP earlier period. but large amount of application of atropine again and again long-term can cause diaphragm function obstacle that is similar to IMS, cause the respiratory failure. The pralidoxime chloride can't counterwork the function of M-receptor (MR),N-receptor (NR) of the nerve center because of passing the blood brain cover, The high quantity of pralidoxime chlorid can restrain the acetylcholinesterase (AchE) and conduce neuro-muscular junction (NMJ) dysfunction of respiratory muscle, thus conduces respiratory muscle to restrain. Be showed from this, atropine and pralidoxime chloride haven't the obvious treatment result to RMP conduced by AOPP. Penehyclidine Hydrochloride can counterworks the function of MR,NR of the nerve center and MR, NR of outsides, the function is stronger that compared with atropine. At the present time, Penehyclidine Hydrochloride has already extensive been applied in AOPP salvage and treatments, and has obtained good of curative effect. But currently, the aspects of reports if this medicine can cure the RMP conduced by AOPP are less. This text discussed and studied if Penehyclidine Hydrochloride can protect diaphragm and the pathogeny of RMP . Experiment Methods: The number of Wistar rats was totally 96, among them 24 for A experiment and 72 for B experiment. The rats was divided randomly into four groups in average: the brine matched control group (NO group); the omethoate intoxication matched control group (PO group); omethoate intoxication and atropine cure group (AC group); omethoate intoxication and penehyclidime hydrochloride cure goup (PC group). The number of rats in each group was 6 in A experiment, 18 in B experiment. And then each group in B experiment was divide evenly into 24 hours team, 72 hours team and 7 days team withrandom, and the number of rats in each team was 6. The poisoned model was made by omethoate (80mg/kg) eliac injection. The respiratory rate (RR) of the anesthetized rats were monitored by BL-410 experiment system in A experiment before giving medicine and 10minutes, 30minutes, 60minutes and 90minutes later. The blood samples were taken for detection of Creatine Kinase(CK)and Cholinesterase (ChE) two hours after giving medicine. In B experiment, the appear and disappear time of organophosphate poisoning symptom was observed in conscious freely moving rats, and the diaphragm specimen was taken for histological confirmation at 24 hours, 72 hours and 7 days after giving medicine. Experiment Results: 1. clinical behave variety of each group rats. The rats in NO group moved freely and had no symptom. The saliva, fasciculation, dyspnea and miosis signs of rats appeard early and the performance was heavy in the PO group, all of them in PO group died in 30-120 min after poisoning; The saliva and miosis signs of rats weren't observed in AC group, the fasciculation signs were alleviated gradually, the respiration was short big in early period and changed gradually to slowly and shallow, 4rats were dead in AC group in B experiment, and one of them died about 24 hours later; The saliva and miosis signs of rats weren't notable in PC group, the fasciculation signs of rats is slight, the active recovering time of rats...
