Subjects: (1)Approaching about hereditable toxicant effect on the occupational workers exposed to low-dose TCE within long term ;(2) Observing the process that how cow-bezoar or bilirubin effect on the DNA damage in lymphocyte and Lipid peroxidation of the occupational workers exposed to low-dose TCE, discussing the anti-oxidation of cow-bezoar and bilirubin , and searching for a suitable toxin-antagonist for TCE intoxication; (3) Analysing the dosage-reaction correlation between ICR mouse' s DNA damage made by cow-bezoar or bilirubin' s antagonism for TCE and Lipid peroxidation, and trying to find the suitable dose for cow-bezoar and bilirubin as toxin-antagonist for TCE.Material and methods: (1)Exposure group: 30 workers exposed continuously to low density of TCE over 3 months; Control group: 30 persons for routine out-patient clinic health-examination. Refering to kit of Nanjing Jiancheng biology research institute, the content or activity of MDA, ROS, SOD and GSH-Px in venous blood were detected;At the same time the DNA breakage of peri-blood lymphocyte (take ratio of movement, Oliver tail moment, length of tail, tail/head (extent), tail/head (inertia), comet extent and comet distributed moment as the effect indication for DNA damage )were detected also; (2)Dealing with peri-blood lymphocyte and peri-blood of the occupational workers exposed to low-dose TCE with 10ymol/L of cow-bezoar or bilirubin, then comparing the level difference of lymphocyte' s DNA damage and plasma lipid peroxidation among 3 groups. (3) Definiting the modeling occupational contact dose according to the DNA damage comet rate (about 80%), and feeding 20, 100, 200 Mmol/L bilirubin or cow-bezoar to corresponding dose poison mice, then observing the antagonist effect ofbilirubin or cow-bezoar in the range from 20 to 200ujnol/L on DNA damage and lipid peroxidation of ICR mice.Results: (1) The lipid peroxidation and cell hereditable toxicity of low-dose TCE acting on the occupational workers: Cj)The SOD activity of exposure group showed an significant descent compared with control group (p<0.01) , whereas the GSH-Px activity showed an significant rise ( p SODn CAT increase significantly except content of MDA . (2)Cow-bazoar or bilirubin effecting on DNA damage of liver , kidney , peri-blood of mice poison by 1706mg/kg TCE: decline of the indications compared with control in both group when bilirubin and cow-bazoar showed various in the range of 20 ~ 200Mmol/L dose (p<0. 05) ; For bilirubin in the range of 20~200Mmol/L, each indication reduced firstly, then increased with the increase of concentrate, which reflected the damage of cell turned weak then strong, the most weaken till lOOMmol/L.Conclusions: (l)The TCE concentration in the air within national safety range showed a effect of lipid peroxidation on workers. (2)Concentration of 10 M mol/L of bilirubin and cow-bazoar can protect DNA from damage through inhibition of lipid peroxidation of the occupational workers exposed to low-dose TCE. (3)Bilirubin and cow-bazoar in the range of 20 ~ lOOMmol/L can strengthen the anti-oxidation of ICR mice, antagonize the DNA damage of liver, kidney, peri-blood cell of mice caused by TCE.
