Fructus Mori Albae, one of the Traditional Chinese Medicine is made by mature fruits of mulberry. According to the Pharmacopoeia, Fructus Mori Albae is usually used as an adjuvant and its medicinal value needs to be further enriched. First the various functional components of mulberry were analyzed and the results showed that the contents of total flavonoids, polypHenols, anthocyanin Pigment and polysaccharides accounted for 0.56% dry weight,2.26% dry weight,0.086% dry weight and 8.62% dry weight respectively. Then this paper analyzed the components of flavonoid aglycones in mulberry by HPLC. The results showed that the contents of quercetin and kaempferol were 1.284 mg and 0.248 mg per gram.The method of extraction of mulberry flavonoids is as follows. First the mulberry was dried to constant weight, then crushed through 60 mesh plug, defatted with petroleum ether, extracted with ethanol, after recovering the ethanol, re-melted in hot water, and extracted with ethylacetate.The last step was to recover the ethylacetate, then the Ethylacetate Extract of Mulberry (EEM) was gained.This paper studied the biological activities of EEM and MFV in vitro. The antioxidant activities of EEM was evaluated by determining the scavenging ability on DPPH·and·OH. The cancer prevention ability of EEM was evaluated by exploring its scavenging nitrite and blocking synthesis of nitrosamine effects. The inhibition effects of EEM were tested on a-glucosidase and xanthine oxidase in vitro. It was found that the scavenging effect of these two kinds of powders on·OH was much stronger than that on DPPH·, and EEM could remove nitrite and block nitrosamine synthesis effectively. What's more, EEM not only had obviously inhibited effects on a-glucosidase, but also had significantly inhibited effects on xanthine oxidase.This study had proved that EEM had obviously inhibited intestinal a-glucosidase activity of rats, so further study was focused on the effects of EEM and its effect on blood glucose. This study had chosen the field of the effects of EEM on glucose metabolism in STZ-induced diabetic rats. The type 1 diabetic rats induced by STZ were randomLy divided into 5 groups, given intragastrically water, glibenclamide and three different level of EEM respectively once a day for 3 weeks, and measured the glucose level per week. The results indicated that EEM showed negative effects on raising of blood glucose and glycosylated serum protein in diabetic rats. In addition, the weight loss was alleviated and the impaired glucose tolerance was significantly improved by oral administration of EEM in diabetic rats. More importantly, EEM at the dose of 100 mg/kg body weight could increase serum insulin level in diabetic rats, which suggested that EEM could improve the abnormal glucose metabolism in STZ-induced diabetic rats.After three weeks' treatment of hyperuricemia rats, the results showed that, three doses of EEM 100 mg/kg,200 mg/kg.300 mg/kg body weight could significantly reduced the level of uric acid, EEM could also obviously reduce the level of urea nitrogen and Cr in rats, which indicated that EEM had a positive effect on kidneys. Furthermore, the microstructures of kidney cells were observered with transmission electron microscopy, which showed that EEM could increased the quantity of mitochondria repaired the shape of karyon and mitochondria. Contrasting with kidney cells of rats, AP treated group showed, karyon irregular, nuclear pore blur, and mitochondria decreased. This also indicated that EEM had positive influence on kidney of hyperuricemia rats.Then the research focused on the extraction process of MFV, and made a flow chart of MFV preparation process. According to the chart, we analyzed the energy consumpution. Next we analysed the activities of MFV in vitro, and the results showed that, the MFV had similar features with EEM, showed good antioxidant activity and good a-glucosidase enzyme and xanthine oxidase inhibition.In summary, the main components of flavonoid aglycones in hydrolysis of EEM and MFV were quercetin and kaempferol. Both have significant antioxidant activity, and obviously inhibition ability of a-glucosidase and xanthine oxidase in vitro. In addition, the EEM could significantly reduce glucose metabolism in STZ-induced diabetic rats, and could also reduce the level of uric acid of hyperuricemia in rats.
