Methods The PH value was detected during the degradation of PDLLA/HA and the morphological characters, cell proliferation, and alkaline phosphatase activity were detected when BMSCs were cultured with PDLLA/HA in vitro. Results During degradation, the PH value of PDLLA/HA decreased little within 12 weeks. BMSCs could be attached to and extended on PDLLA/HA, the proliferation and growth were not affected by PDLLA/HA. Methods The composite of BMSCs and PDLLA/HA was implanted in subcutaneous sites in rabbits. All implants were examined by gross observation andunderwent histological examination. Results Cartilage and bone are observed in the composites of the BMSCs and PDLLA/HA material in rabbit models. There was not any cartilage and bone formed at control group. PDLLA/HA could be used as a scalfold of bone tissue engineering.Part III PDLLA/HA combined with bone marrow stromal cells for repairing radial defects in rabbitsObjective To study the defects of combined use of PDLLA/HA and BMSCs on repairing segmental bone defects. Methods 10mm radius defect was made in 72 forelimbs of 36 adult New Zealand white rabbits. The 72 forelimbs were randomized into 3 groups, the defects of group A were filled with BMSCs-PDLLA/HA, the defects of group B were treated with PDLLA/HA, group C was untreated(empty) to serve as the control group. The healing response was evaluated macroscopically, histologically and radiographically at two, four, eight, and twelve weeks after implantation. Results Group A exhibited the strongest defect-repairing ability ,and obvious healing of could be seen 12 weeks postoperatively.At every stage, Group A did a better job than Group B in terms of quantity and quality of new bone growth. The score of x-ray in the implants loaded with BMSCs was higher than did cell-free implants.(p<0.01) Conclusions Since the BMSCs- PDLLA/HA can repair segmental bone defects, it can be used in the clinical treatment of bone defects.
