Objective To investigate the correlation between HnRNPA2/Bl,HnRNPBl overexpression and lung cancer development and to evaluate their value in the early detection for human lung cancer.Methods HnRNPA2/B1 mRNA and HnRNPBl mRNA were separately studied in lung cancerous tissues and paracancerous tissues by RT-PCR.We analyzed and compared the difference and the changing tendency between them in all TNM stages and histological types of tumors.Results①The positive rate(79.1%) of HnRNPA2/B1 in cancerous tissues was significantly higher than that in paracancerous tissues(33.3%)(P<0.01).And HnRNPBl was 81.4% to 40%(P<0.01).ⅢThere was a tendency that the later TNM stage was,the less expression rates of HnRNPA2/B1 and HnRNPBl were.They had greater expression in the preneoplastic lesions and early stage tumors,and decreased in advanced tumors.③ HnRNPA2/B1 and HnRNPBl were found less frequently in adenocarcinoma compared with others histological types. There was no significant association between HnRNPB1; mRNA level and histological types of tumors.④We analyzed the level of both HnRNPA2/Bl mRNA and HnRNPBl mRNA isolated from cancerous(T) and noncancerous(N) tissues. HnRNPA2/Bl mRNA was equally expressed in the cancerous and noncancerous tissues (T/N=0.9~1.5). However, HnRNPBl mRNA was especially ealevated in cancerous tissues compared with the level in noncancerous tissues (T/N=l .5~7.34).The level of HnRNPBl mRNA gradually decreased with the stage advancing.There was a significant higher level in stage I tissues (5.94+2.11) than that in stage II -IV (2.36±1.99) (P/><0.05) .Conclusions HnRNPA2/B1 and HnRNPBl had significant overexpression in both preneoplastic lesions and tumors.The higher expression was found in earlier stage tissues. The overexpression of HnRNPA2/Bl and HnRNPBl is an early event in oncogenesis,which could be sensitive markers for the early detection for lung cancer.
