Objective: Lung cancer occupied a major position in the causes of death fromcancer, with the mortality and morbidity presenting a rising trend. However, the earlydiagnosis of lung cancer was still a unsolved problem, of which only2%-3%couldobtained an early diganosis. Non-small cell lung cancer took a percentage of80-85%in all kinds of lung cancer. The studies on protein expression related lung cancer hadalways been the hot spot of the lung cancer research field. It was showed thathnRNPA2/B1expressed at a high level in non-small cell lung cancer, and it was alsoproved that it was increased in the peripheral blood of patients with NSCLC than withnormal control and benign lung disease by using real-time fluorescence quantitativeRT-PCR. However, the study to confirm hnRNPA2/B expressing in peripheral bloodby immunohistochemical method has been seldomly reported.This study respectively detected hnRNPA2/B1expressions in the sputum andblood of patients with NSCLC, lung benign lesions and normal controls by usingimmunohistochemical method, and explored the clinical correlation between NSCLCand hnRNPA2/B1(including age, gender, smoking situation and pathologicalcategory).Methods: Participants in this study included162patients with NSCLC,30patients with benign lung disease and20normal controls, who were hospitalizedin the first affiliated hospital of Nanchang Uiversity from September2010to May2011. The pathologies of all cases expcept healthy subjects were made a definitediagnosis by transthoracic needle biopsy、bronchoscopy with biopsy or surgicalresection pathology, and meanwhile these cases had not ever received anyradiotherapy or chemotherapy. Then the associated dates were statistically analysedby using SPSS17.0software.Results:1、The expression of hnRNPA2/B1in lung cancer group:the number of caseswhose hnRNPA2/B1showed positive expression was36(36/60, the postive rate60%) in the sputum,and48(48/60, the postive rate80%) in the blood respectively.2、The expression of hnRNPA2/B1in benign lung disease group:the number ofcases whose hnRNPA2/B1showed positive expression was11(11/30, the postive rate36%) in the sputum,and9(9/30, the postive rate30%) in the blood respectively.3、The expression of hnRNPA2/B1in normal control group:the expression ofhnRNPA2/B1in the blood was positive in19healthy subjects,while that wasnegative in1healthy subject,with the postive rate5%.4、It was judged positive rate by10%to detect hnRNPA2/B1expression in theblood by immunohistochemical method.5、The expression of hnRNPA2/B1in the blood and sputum showed nocorrelation with gender、age、smoking situation and pathologic type.6、The sensitivity and the specificity of detecting hnRNPA2/B1expression were80%and70%in the blood respectively,while60%and63%in the sputumrespectively.Conclusion:Detecting hnRNPA2/B1in the blood and sputum by immunohistochemicalmethod had a certain diagnostic value in early diagnosis of lung cancer.Dectecting the expression of hnRNPA2/B1in the blood cells byimmunohistochemical method had a important value in diagnosis and differentialdiagnosis of lung cancer,the efficiency of which was higher than it of dectecting inthe sputum.The expression of hnRNPA2/B1was not apparently related to gender、age、smoking situation and pathologic type.
