Objections: To examine main chemical constituents and influence to model rats' urinary catecholamines excretion of HuangLian-EJiao decoction. Methods: Having utilizing modern analytical technology including GC-MS, HPLC and HPLC-ESI-MS, to study chemical constituents of HuangLian, HuangQin, BaiShao's whole .decoction and the influence of different cooperation methods to effective compounds in the supernatant. In addition, having researched the hydrolysis of the whole decoction's deposition by artificial gastric juice and artificial intestinal juice. At the same time, having built up the model of high urinary catecholamines excretion by i.p. ACTH to SD rats, then the model was seperated to six groups, negative control group, contrastive medicine group, low dosage group, middle dosage group, high dosage group, alternative medicine group, and i.g. distilled water, Tianwangbuxin Wan, low, middle, high dosage's Huanglian-Ejiao decoction and alternative medicine, 2mL/100g weight/day for 15 days, respectively. Results: Identified 19 volatile compounds and the other 15 compounds from petroleum ether, benzene and methanol extraction of the whole decoction in all. It shows that berberine, baicalin and paeoniflorin in the whole decoction's supernatant decreased 72.96%, 81.92% and 9.82%, respectively, compared with the single decoction of HuangLian, HuangQin and BaiShao. Furthermore, after hydrolysis, both berberine and baicalin was detected but paeoniflorin. Nevertheless, compared with the single decoction, 62.14 percents of berberine and 70.91 percents of baicalin were losing yet. After i.p. ACTH for 4 days, average volumes of rat urine has increased 43.15%, besides urinary norepinephrin, epinephrin, dopamin excretion have increased 17.85%, 49.90%, and 5.14%, respectively, and that SD rats appeared some behavior's changes including excitement, activity-increasing, irascibility, difficulty to snatch. During the oral administration, volumes of urine have showed different trend, and, urinary catecholamine excretion of each group have decreased on different degree. After administration for 15 days, average volumes of urine of middle dosage group decreased 20.44%, urinaryepinephrin, dopamin excretion decreased 41.76%, 9.13%, respectively, markedly highly to negative control group and other groups. Conclusions: There are a small quantity of volatile compounds and HuangLian alkaloids, HuangQin fiavones, as well as BaiShao monoterpenoids in the whole decoction. Compared with the single decoction, much of berberine and baicalin were losing during decocting wholly, and, even though the deposition have been hydrolyzed, only were small part of berberine and baicalin detected, consequently, writer suggest that it should be the better way that decocting the raw plant materials solely and then mixing the extraction. Rats model have showed self-cure ability, Tianwangbuxin Wan could inhibit that ability, and middle dosage HuangLian-EJiao decoction have the best effect, at the same time, compared with original prescription, the alternative medicine's effect was highly weaken. HuangLian-EJiao decoction has showed the well curative effect to rats model, simultaneously, which, in turn, has demonstrated that pathological changes of SD rats model by i.p. ACTH could be similar to YinXuHuoWang Zheng of Chinese Medicine, partly.
