[Objective] To identify the single nucleotide polymorphism (SNPs) of angiotensin II type 2 receptor (AGTR2) gene and to determine whether the AGTR2 gene SNPs are associated with essential hypertension (EH) in a male Chinese population in Shandong Province.[ Methods ] Genomic deoxyribonucleic acid( DNA) was extracted from 96 male of essential hypertension and 119 healthy control subjects from peripheral blood leukocytes by standard method. The detection of SNPs was performed in 20 subjects by direct DNA sequencing. 96 male patients with EH and 107 normal blood pressure controls were included in the study to assess the contribution of SNPs of AGTR2 gene to hypertension in men.[Results] Seven SNPs of AGTR2 gene were detected among which 2 SNPs were reported for the first time.Two SNPs are in promoter, one in intron, one in encoding region, three in 3'UTR. A case-control study using two SNPs ( A1675G and T1334C ) showed that there is no different in allele frequency of C1334 between the male hypertensive subjects and healthy control subjects.(16.7% vs 11.2%, P>0.05), but there is a significant increase in allele frequency of A1675 in male hypertensive subjects(49.0% vs 34.6% for normotensive subjects, PO.05).[ Conclusion ] SNP A1675G of AGTR2 gene might be involved in the development of essential hypertension in male Chinese.
