CA is a sexually transmitted disease, which is caused by the pathogen—Humanpapillomavirus (HPV), to infect the only hosts-human beings, and is characterized bybenign verrucous hyperplasia on genitals, crissum and so on. The most patients of CAare usually young and middle-aged people, who infect each other mainly throughsexual contact, rarely through indirect contact.CA has these features of beinghyperplastic fast and being recurrent easily; and may affect seriously daily life of thepatients, which will be harmful to society and families without doubt. Therefore, CAhas been considered as one of the most important monitoring venereal diseases.XIAP is one of the most effective profilin in the endogenous caspase pathwayamong inhibitor of apoptosis proteins (IAPs). XIAP has the ability of blocking cellapoptosis by the way of combining apoptosis promoter (caspase-9) and apoptosiseffective factors (caspase-7and caspase-3).Many research indicate that XIAP ishighly expressed in most of malignant tumors and also plays an important role in theprocess of cell proliferation and the process of neoplastic transformation; C-jun is aproduct, which is generated from expression of jun genes and can code the elementsof transcription factor AP-1(activator protein-1) induced by TPA (TissuePolypeptide Antigen);After c-jun is combined with the family of fos proteins(including c-fos,fosB,Fra-1and Fra-2) into homodimeric or heterodimer, then, c-juncan regulate cell growth and differentiation through combination with TPA responseelements in various promoters of genes.CA is a kind of benign hyperplastic disease, and however, it also probably develop into malignant tumor though the rate is low. Toa certain extent, it shows that CA tissues may exist abnormal expression ofapoptosis-related genes. In recent years, the research on the relationship between CAand cell apoptosis are often carried out, however, there are yet no reports about theexpression of apoptosis-related factors, XIAP and c-jun in CA tissue. In this test, weadopted immunohistochemical SABC method to investigate the expression and thesignificance of XIAP and c-jun in CA and thereby explore further their possible effecton the pathogenesis of CA.Material and Methods1Objects of the studyForty specimens of CA were collected from outpatients in the department ofdermatology and venereology from September2012to June2013;The cases of all thepatients, who had typical clinical manifestations, and did not any other diseases, andalso did not accept any treatment before seeing a doctor, conformed to the diagnosticcriteria of CA; There were28males and12females, whose ages ranged from18years old to59years old (mean28.4士10.3years old);18specimens as the controlgroup, were obtained from the normal foreskin tissues of adult males, whounderwent surgical operations in the first affiliated hospital of Zhengzhou university.2MethodsWe adopted SABC immunohistochemistry method to measure the expression ofXIAP and c-jun in the40cases of CA tissues and18cases of normal prepuce tissuesas control. The procedures shall be carried out in accordance with kit instructions.According to the authoritative reference, semi-quantitative ranking score was appliedaccording to the positive cells percentage and color intensity. The score would be0point if the positive cells percentage was10%or less, one point if11%~25%,twopoints if26%~50%, three points if51%~75%,four points if76%or more. The scorewas0point if the color intensity was negative,1point if buff,2points if yellow, and3points if ocher yellow. On the basis of the above score ’s respective multiplication, the final score of the expression of XIAP and c-jun in CA could be gotted (0point,negative;1point~4points, weakly positive;6points~8points, positive;9points~12points, strongly positive).3Statistical analysisBy using SPSS17.0statistical software, Chi-square test was used to test thecomparison of positive rate,and Spearman test was used to analyze the correlationbetween XIAP and c-jun.4Results1. The expression of XIAP in CA and normal foreskin.The positive expression of XIAP in CA, localized in cytomembrane or cytoplasm,was stained buffy or darkbrown and was observed in mostly stratum spinosum of thewhole epidermal layers. In40cases of CA, the expression of32cases showedpositive; The positive expression rate was80%, and the intensity of positiveexpression was++~+++. In18cases of normal skins, the expression of5casesshowed positive; The positive expression rate was27.8%, and the intensity ofpositive expression was-~++.There was statistical significance of the positiveexpression rate and the positive expression intensity between the two groups(P<0.001).2. The expression of c-jun in CA and normal foreskin.The positive expression of c-jun in CA, which was mainly localized in cell nucleus,was stained buffy or darkbrown and was observed in mostly every layer of the wholeepidermal layers without stratum corneum. In40cases of CA, the expression of36cases showed positive; The positive expression rate was90%, and the intensity ofpositive expression was++~+++. In18cases of normal skins, the expression of3cases showed positive; The positive expression rate was16.7%, and the intensity ofpositive expression was-~++. There was statistical significance of the positiveexpression rate and the positive expression intensity between the two groups(P<0.001).3. The correlation analysis between the expression of XIAP and c-jun in CA. In32cases of positive XIAP, there were30cases of positive c-jun and2cases ofnegative c-jun. In36cases of positive c-jun, there were30cases of positive XIAP and6cases of negative XIAP. By the method of Spearman rank correlation coefficienttest, we found that the expression of XIAP was positively correlated with c-jun in CA(P=0.004).Conclusion1. The positive expression rate of XIAP in CA was significantly higher than that incontrol group. Suggesting that over-expression of XIAP could promote cellproliferation by the means of inhibiting the cell apoptosis.2. The positive expression rate of c-jun in CA was significantly higher than that incontrol group. Suggesting that over-expression of c-jun may be involved withabnormal proliferation of cells in CA.3.The expression of XIAP was positively correlated with c-jun in CA tissues,suggesting that both of the proteins corporately influenced the pathogenesis andrecurrence mechanism of CA....
