Study On The Effect Of Total GL Of Chishao On The Level Of CK And Apoptosis Of The Tumor Cell In The Tumor-Bearing Mice

Object: To study that TGC was acted as in the tumor mice of S₁₈₀, TGC have an important influence on inducing the tumor cell apoptosis and secreting cytokine. Further perfect TGC the anti tumor function mechanism. Method: After built the tumor mold The ratio of antitumor: Weigh the intact tumor of group and calculate the ratio .The appearance learns the observation: The light microscope, the electricity mirror observes the each tumor organization pathology characteristics. Observing TGC act on tumor mice, the mRNA transcription of c-myc were evaluated in situ hybridization and the protein expression of Bcl-2 and P16 were evaluated by SABC immunohistochemistry. IL-10 was measured by the ELISA method, while we can observe the influence of the TGC acting on the tumor cell of cytokine. Result:TGC group , TGC+CY group and CY group comparing with model group show the difference of significance in the ratio of antitumor. Observation of Tumor organization the pathology show that TGC has obvious the anti-tumor function,TGC group and model group show the difference of markedness in the positive expression of Bcl-2, c- myc and P16 . Bcl-2 has a function to repress the cell apoptosis, c-myc is oncogene and controls the cell propagate, P16 is tumor suppressor gene, TGC group has presented to descend the trend that Bcl-2 and c-myc express to descend and P16 express to rise. IL-10 is negative regulate factor, TGC can decrease the level of IL-10. Conclusion: The TGC has regulated cytokine to drift the phenomenons of the Th1/ Th2 in tumor body , maintain Th1 of advantage appearance and regulate the immunity function of the tumor mice.TGC regulate the expression of Bcl-2, c-myc, P16 gene in order to not only apoptosis the tumor cell but also inhibit tumor cell proliferation .The experiment has proved that TGC has the obvious antitumor function.
